摘　　要：目的探讨单纯疱疹病毒1型(HSV-1)对星形胶质细胞内胞核转录因子kappa B(NF-κB)转录活性及炎性细胞因子肿瘤坏死因子α(TNF-α)和白细胞介素10(IL-10)表达的影响。方法 HSV-1感染星形胶质细胞,通过ELISA,Western blot等方法检测星形胶质细胞NF-κB,TNF-α,IL-10的表达;同时研究NF-κB通路抑制剂吡咯烷二硫氨基甲酸(PDTC)对TNF-α,IL-10表达的影响。结果星形胶质细胞被HSV-1感染后,胞核内NF-κB蛋白表达明显增强,星形胶质细胞分泌的TNF-α,IL-10均上调,与对照组比较差异有显著性(P<0.05)。PDTC可抑制NF-κB活化,导致核内NF-κB蛋白表达下调,使得细胞分泌TNF-α,IL-10降低,与HSV-1组比较差异显著(P<0.05)。结论被HSV-1感染的星形胶质细胞通过活化NF-κB,从而上调TNF-α,IL-10的表达。Objective To investigate the effects of herpes simplex virus type 1 （ HSV-1 ） on expression of nuclear transcription factor kappa B（NF-κB）,the cytokines of tumor necrosis factor alpha（TNF-α） and interleukin-10 （IL-10） in astrocytes.Methods HSV-1 infected astrocytes and proteins of NF-κB,TNF-αand IL-10 were detected by Elisa and Western blot respectively.The effects of NF-κB inhibitor pyrrodlidine dthiocarbamate（ PDTC） on expressions of TNF-αand IL-10 were also detected.Results Compared with the control group,when infected by HSV-1,the expres-sion of nuclear NF-κB,TNF-αand IL-10 were all up-regulated,the difference between them was significant（ P〈0.05） . The PDTC inhibited NF-κB activation and resulted in down-regulating protein expression of nuclear NF-κB,which lead to down-regulation of TNF-αand IL-10,compared with the HSV-1 group,the difference was significant（P〈0.05）.Con-clusion Astrocytes infected by HSV-1 can activate NF-κB so as to up-regulate the expression of TNF-αand IL-10.
