摘 要:目的 研究姜黄素体外对人结肠癌Lovo细胞血管内皮生长因子VEGF表达的影响。方法不同浓度(0μmol/L、5μmol/L、10μmol/L、20μmol/L、40μmol/L)姜黄素处理2013年8月—2014年2月中南大学提供的人结肠癌细胞株Lovo24 h后,用Real-time PCR检测姜黄素对人结肠癌细胞株血管内皮生长因子VEGF m RNA表达的变化,用Western blot法检测姜黄素对人结肠癌细胞株VEGF蛋白表达的变化。结果人结肠癌Lovo细胞经过姜黄素处理24 h后,5μmol/L、10μmol/L、20μmol/L、40μmol/L姜黄素组VEGF m RNA及蛋白的表达均明显下降,与对照组比较差异有统计学意义(P<0.05)。结论姜黄素能下调人结肠癌Lovo细胞VEGF的表达,并呈剂量依赖,这可能是姜黄素抑制结肠癌Lovo细胞增殖及侵袭性的机制之一。Objective To study the effect of curcumin on the expression of VEGF in human colon cancer Lovo cells in vitro.Methods The human colon cancer cell lines Lovo were treated with 0 μmol/L,5 μmol/L10 μmol/L,20 μmol/L, 40 μmol/L curcumin for 24 hours respectively, then we detected the human colon cancer cell lines VEGF m RNA expression by Real-time PCR and VEGF protein expression by Western Blot. Results After the human colon cancer cell lines Lovo cells treated with curcumin for 24 hours respectively, the expression of VEGF m RNA and protein of the curcumin-treated groups(5 μmol/L,10 μmol/L,20 μmol/L,40 μmol/L) had more significant reduction compared with control group(P0.05). Conclusion Curcumin could decrease the expression of VEGF in human colon cancer Lovo cells dose-dependently, this may provide some evidence for the mechanism that curcumin could inhibit the proliferation and invasion of colon cancer Lovo cells.
