摘 要:目的探索香附油滴丸抗痛经作用及其机制。方法采用小鼠离体子宫平滑肌收缩试验,观察受试药对子宫平滑肌收缩张力及p42/44丝裂原激活蛋白激酶(p42/44MAPK)磷酸化水平和间隙连接蛋白43(Cx43)表达水平的影响;采用缩宫素致小鼠痛经试验,观察受试药的抗痛经作用及对血栓烷B2(TXB2)和6-酮-前列腺素F1α(6-Keto-PGF1α)表达、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活力的影响。结果受试药连续灌胃给药7 d,显著降低小鼠子宫平滑肌收缩张力及p42/44MAPK磷酸化水平和Cx43表达水平(P<0.05,P<0.01);明显抑制缩宫素致小鼠痛经反应,降低小鼠血浆TXB2水平和血清MDA含量及升高小鼠血浆6-Keto-PGF1α水平和血清SOD活力(P<0.05,P<0.01)。结论香附油滴丸具有明显的抗痛经作用,其作用机制可能与抑制p42/44MAPK磷酸化水平和Cx43表达,调节TXB2/6-Keto-PGF1α平衡及抗氧化作用相关。Objective To explore the anti-dysmenorrheal effects and mechanism of Xiangfu Y ou Diwan(XYD). Methods The isolated mouse uterine smooth muscle contraction test was carried out to explore the action of XYD on tension of uterine smooth muscle,p42/44 mitogen-activated protein kinase (p42/44MAPK)activity and connexin 43 (Cx43) expression. The oxytocin- induced dysmenorrhea test in mouse model was used to evaluate the anti-dysmenorrheal effect of XYD, the expression levels of thromboxane B2 (TXB2) and 6-keto-prostaglandin F1α(6-Keto-PGF1α),malondialdehyde(MDA)content,and superoxide dismutase(SOD) activity in blood. Results XYD significantly decreased the contraction of mouse uterine smooth muscle, p42/44MAPK activity and Cx43 expression after oral administration for 7 days (P〈0.05,P〈0.01),and obviously relieved the oxytocin-induced dysmenorrhea, decreased TXB2 level and MDA content,and increased 6-Keto-PGF1α level and SOD activity in blood(P〈0.05,P〈0.01). Conclusion XYD has significant anti-dysmenorrheal effects, and its mechanism may be relevant to inhibiting p42/44MAPK activity and Cx43 expression, regulating the balance of TXB2, 6-Keto-PGF1α and counteracting oxidation.
