摘 要:目的正交试验结合药效学实验优选黄芪葛根汤抗乙醇诱导胃黏膜损伤最佳水煎工艺。方法小鼠灌胃给药4 d后,无水乙醇灌胃制备小鼠急性胃黏膜损伤模型,以胃黏膜损伤分数作为评价黄芪葛根汤保护胃黏膜的药效学指标。对黄芪葛根汤水煎液的煎煮时间、煎煮次数、加水量3个因素进行正交试验设计,以葛根素的含量作为评价指标,优选最佳水煎工艺。结果黄芪葛根汤抗胃黏膜损伤的最佳提取水煎工艺为煎煮30 min,煎煮3次,加10倍水。结论:正交试验优选的黄芪葛根汤水煎工艺简便可行,可充分提取汤剂中葛根素,并起到保护胃黏膜的作用。Objective To optimize the water extraction process of Huangqi Gegen decoction (HGD) in protecting gastric mucosal injury by orthogonal test coupled with pharmacodynamics experiment. Methods After the mice were given HGD orally for 4 days,absolute alcohol was given to the mice intragastrically to induce the acute gastric mucosal injury model . Gastric mucosal injury index was calculated to study the pharmacodymamic effects. With the content of puerarin as the observation index, and using the boiling time, boiling frequency, the amount of water as factors, water extraction process of HGD was optimized. Results The optimal condition of water extraction process of HGD in protecting gastric mucosal injury was as follows:adding 10-fold water to decoct for 3 times, 30 min for each time. Conclusion The water extraction process of HGD optimized by orthogonal test is simple and reliable, and it could thoroughly extract puerarin from the decoction to protect gastric mucosa.
