摘  要:目的:研究戊巴比妥钠在2型糖尿病大鼠模型复制中的应用效果。方法:雄性Wistar大鼠,实验组采用"高脂饲养+戊巴比妥钠+链脲佐菌素(streptozotocin,STZ)"的方法复制2型糖尿病模型,对照组采用"高脂饲养+STZ"的方法复制模型,各时期以空腹血糖>7.8mmol·L-1作为2型糖尿病模型的判断标准。结果:(1)实验组大鼠胰腺与肝脏组织呈现典型的糖尿病病理改变;(2)STZ注射后第2、4、6、8周比较,实验组大鼠成模率都显著高于对照组(P<0.01);(3)在STZ注射后第6和8周,实验组死亡率显著低于对照组(P<0.01);(4)实验各期血糖变异度比较,实验组大鼠显著低于对照组(P<0.01)。结论:戊巴比妥钠结合小剂量STZ可以提高2型糖尿病模型的成功率和稳定性,降低死亡率和模型之间的血糖差异性。Objective:To study the application effects of pentobarbital sodium in rats model of type 2 diabetes mellitus(T2DM). Meth-ods:Male Wistar rats were used in the experiment. The experimental group rats were used in the“high-fat chow + pentobarbital sodium+ streptozotocin( STZ)”to copy the model of type 2 diabetes. The control group rats were used in the“high-fat chow + STZ”to copy the model. Rats with fasting blood glucose〉7. 8 mmol·L-1 were taken as standard of T2DM. Results:(1)The experimental group rats’ pan-creas and livers tissue showed changes in diabetic pathological type;(2)After injecting STZ,compared the 2nd,4th,6th and 8th week,the success rate of the experimental group is significantly higher than that of the control group(P〈0. 01);(3)The 6th and 8th week,the death rate of the experimental group is significantly lower than the control group(P〈0. 01);(4)The glucose variability of the experimental group is significantly lower than those in the control group(P〈0. 01). Conclusion:Pentobarbital sodium combined with low doses of STZ can improve the success rate and stability of model of type 2 diabetes and reduce the mortality and glucose variability.
