摘  要:目的建立高效液相色谱法测定人血清中维生素A的方法。方法血清加入维生素A醋酸酯内标,以乙醇沉淀蛋白质,用正己烷提取维生素A,氮吹浓缩后采用HPLC紫外检测器测定。结果维生素A的线性范围为0.05~2.0μg/ml,标准曲线的相关系数大于0.9995,方法回收率在92%~111%之间,日内日间相对标准偏差(RSD)小于6%。结论该法灵敏、准确、快速,适用于人血清中维生素A浓度的测定。Objective The study aimed to establish a method for determination of vitamin A level in human serum by high-performance liquid chromatography (HPLC). Methods Retinyl acetate was added to the serum samples as an internal standard, and ethanol was used for protein precipitation, n-hexane was used to extract vitamin A from the samples, which were then concentrated under nitrogen and ran through a HPLC UV detector. Results Vitamin A was linear under 0.05-2.0 μg/ml; correlation coefficient of the standard curve was higher than 0.9995; recovery rate of the method was 92-111%; and the same-day relative standard deviation (RSD) was smaller than 6%. Conclusion The method is sensitive, accurate, fast, and is thus suitable for the determination of vitamin A level in human serum.
