摘 要:目的研究阴离子阿奇霉素泡囊(AMNS)的大鼠在体肠吸收性质。方法采用薄膜分散法制备AMNS并测定其粒径与Zeta电位;运用单向灌流模型考察大鼠的十二指肠、空肠、回肠以及结肠的药物吸收情况;采用HPLC法检测药物的浓度,并计算其吸收速率常数(Ka)及有效渗透率(Peff)。结果制备所得AMNS的粒径为1.05μm,Zeta电位为-40.1 m V;AMNS在十二指肠、空肠、回肠以及结肠中的Ka和Peff分别是阿奇霉素的1.56、1.10、1.27、1.47倍和2.14、1.22、1.31、1.06倍。结论AMNS能明显改善药物在大鼠十二指肠、空肠、回肠以及结肠中的吸收。OBJECTIVE To study whether anion Azithromycin niosomes (AMNS) can improve the intestinal absorption of Azithromycin(AM) in rats. METHODS The film - evaporation method was used to prepare AMNS. The size and Zeta potential of AMNS were determined by photon correlation spectroscopy. The rat single pass intestinal perfusion method was employed to investigate the effects of duodenum, jejunum, ileum and colon on drug' s absorption rate constant and the intestinal apparent permeation coefficients. The concentrations of the drugs were determined by HPLC. RESULTS The size and Zeta potential of AMNS were 1.05 μm and -40. 1 mV,respectively. The absorption rate constant(K ) of AMNS in the duodenum, jejunum, ileum and colon was 1.56, 1.10,1.27 and 1.47 times that of the free AM and effective permeability(Peff) was 2.14,1.22,1.31,1.06 times that of the free AM, respectively. CONCLUSION AMNS can significantly improve the absorption of the drug in the small intestine of rats.
