摘 要:目的以RKO细胞株为增殖活力测试的研究对象,比较CCK-8法与Alamarblue法在检测中波长、孵育时间、细胞密度等方面的差异性。方法取体外培养对数生长期的RKO细胞株,分别比较两者在不同检测波长(380、430、480、530、580、630nm)、不同孵育时间(1、2、3、4、5、6h)、不同细胞密度(2×10^4、5×10^3、1.25×10^3CUF·mL^-1)下所测的光密度值(OD值),并比较两者在相同条件下同-标本重复检测时的平衡性。结果ALamarblue法在不同细胞密度中的最佳检测波长皆为580nm,而CCK-8法的最佳检测波长会随着细胞密度的变化而有所改变;1—6h时,CCK-8法适合的孵育时间在1h以后,Alamarblue为2h以后,且CCK-8法的灵敏度、平衡性都强于Alamarblue法。结论CCK-8法在细胞增殖活力检测中的灵敏性、平衡性、快捷性等都略强于ALamarblue法。OBJECTIVE Taking RKO cell lines as the study object of proliferation activity test, to find out the differences between CCK - 8 method and Alamar blue method by comparing some factors such as the detection wavelength,incubation time and cell density. METHODS RKO cells which were cultured in vitro respectively in logarithmic growth phase was used, with the different detection wavelength(380 nm,430 nm,480 nm,530 nm,580 nm,630 nm) ,the different incubation time (1 h,2 h,3 h,4 h,5 h,6 h) ,the differ- ent cell density(2×10^4,5×10^3,1.25×10^3CUF·mL^-1) optical density. The values(OD) were measured,and compared the balance of both RKO cells which were put in a repeated detection by the same specimen and the same conditions. RESULTS 580 nm was the best detection wavelength in the different cell densities by Alamar blue method,while the best detection wavelength of CCK - 8 method changed with the different cell densities. Within 1 - 6 h, the incubation time was 2 h later by Alamar blue method,while it was 1 h later by CCK -8 method. Both the sensitivity and balance facets ,the CCK- 8 method was comparatively more effective than Alamar blue method. CONCLUSION In this cell proliferation activity detection test, the CCK - 8 method is comparatively more effective than Alamar blue method regarding the sensitivity, balance and efficiency.
