摘 要:目的探讨蛋白酶激活受体2(PAR-2)在"肺合大肠"生理机制中的作用。方法将30只SD大鼠随机分为空白对照组、肺生理性激活组、结肠生理性激活组,每组10只。肺生理性激活组采用雾化吸入1%PAR-2激活剂反肉桂酰-亮-异亮-甘-精-亮-鸟-[酰胺](tc-LIGRLO)溶液建立肺生理性激活大鼠模型;结肠生理性激活组采用结肠内灌入1%tc-LIGRLO溶液建立结肠生理性激活大鼠模型。观察各组大鼠肺与结肠功能、肺与结肠迷走神经放电活动、支气管与结肠平滑肌张力及收缩频率。结果肺生理性激活组碳末推进率、结肠平滑肌张力及收缩频率较空白对照组明显增高,激活后10 min时迷走神经放电活动较激活前降低(P<0.05)。结肠生理性激活组气道阻力和肺动态顺应性较空白对照组明显改善,支气管平滑肌张力及收缩频率明显下降,激活后10、15 min迷走神经放电较激活前升高(P<0.05)。结论通过刺激支气管或结肠PAR-2,可使相合脏腑的功能发生改善,说明PAR-2可能在"肺合大肠"生理机制中起着维持肺与结肠气机通降功能的作用。Objective To study the activity of protease-activated receptor 2( PAR-2) in the physiological mechanism of lung being related to large intestine theory. Methods Totally 30 SD rats were randomly assigned to control group,pulmonary physiological activation( PPA) group,colon physiological activation( CPA) group,10 rats in each. The PPA group inhaled PAR-2 activator,1% trans-cinnamoyl-Leu-Ile-Gly-Arg-Leu-Orn- [amide ]( tcLIGRLO) solution to make PPA rat models; the CPA group were used 1% tc-LIGRLO solution by colon administration to make CPA rat models. The pulmonary and colon functions,firing activity of vagus nerve,muscular tension and contraction frequency of the bronchus and colonic smooth muscle were observed. Results The carbon particle propelling rate,muscular tension and contraction frequency of the colonic smooth muscle in the PPA group was significantly higher than the control group,while the firing activity of vagus nerve reduced 10 min after activation( P〈0. 05). The airway resistance and lung dynamic compliance of the CPA group was better than the control group,with the muscular tension and contraction frequency of the bronchial smooth muscle declining and the firing activity of vagus nerve ascending 10 min and 15 min after activation( P〈0. 05). Conclusion By stimulating PAR-2 on the bronchus or colon,the function of organs related to one another could be improved,which indicated that PAR-2 may act by regulating qi movements of the lung and large intestines in the physiological mechanisms of lung being related to large intestine theory.
