摘 要:目的研究黄芩苷对流感病毒感染小鼠早期肺组织病理的影响,探讨黄芩苷对流感病毒致肺损伤的保护分子机制。方法用流感病毒亚甲型鼠肺适应株A/FM/1/47(H1N1)感染小鼠,建立小鼠流感病毒性肺炎的生物模型,用不同剂量黄芩苷溶液灌胃治疗后,制作小鼠肺组织切片观察HE及改良Masson染色后病理变化,采用实时荧光定量PCR测定肺组织TGF-β1、MMP-9 mRNA表达量;采用Westen blot方法测定肺组织TGF-β1、MMP-9蛋白表达量。结果黄芩苷中、小剂量组均能明显减轻肺组织病变,明显抑制肺组织纤维蛋白的分泌,显著下调肺组织TGF-β1、MMP-9 mRNA及蛋白表达。结论黄芩苷可减轻流感病毒感染致小鼠肺组织的损伤,可能与其抑制TGF-β1、MMP-9基因及蛋白表达相关。Objective This study was conducted to evaluate the effect of baicalin on the pathological changes of mice lung tissue with H1N1 infection and its molecular mechanism. Methods The model of mouse influenza virus pneumonia was prepared. After gavage treatment with different doses of baicalin, HE and modified Masson staining specimen were observed;Fluorescent qPCR was used to assess the mRNA expressions of TGF - β 1 and MMP - 9 ; Western blot analysis was adopted to determine the protein ex- pressions of TGF -β 1 and MMP - 9. Results The experiment demonstrated that baicalin, both middle and small doses can sig- nificantly reduce the lesion of lung tissue, inhibit the secretion of fiber protein in lung tissue; significantly down regulate mRNA and protein expressions of TGF - β1 and MMP - 9. Conclusion Baicalin can reduce the lung injury with influenza virus infec- tion. It may be related to the inhibition of gene and protein expression of TGF - β1 and MMP - 9.
