摘  要:目的氟苯尼考磷脂复合物的最佳工艺优化,并对其进行表征。方法以氟苯尼考与磷脂的复合率为评价指标,采用单因素试验和正交试验设计筛选最优制备工艺,考察了药物与磷脂的投料比、药物浓度、反应时间、反应温度这4个因素对磷脂复合物形成的影响,并利用UV、IR、显微镜等分析技术对复合物结构进行分析。结果确定氟苯尼考磷脂复合物的最佳制备工艺为:四氢呋喃为反应溶剂,药物与磷脂的比例为1:2,在45℃条件下反应2h,该工艺条件得到的磷脂复合物复合率为95%。结论 氟苯尼考磷脂复合物的形成受药物与磷脂的投料比、药物浓度以及反应时间的影响较大,反应温度也有一定的影响,且复合物在形成过程中并未发生化学变化,但与单纯的物理混合物有本质的区别。Objective This study was designed for the preparation offlorfenicol phospholipid complex, seeking for the optimum preparation and characterization of it. Methods The single factor experiment and orthogonal experiment were used to design to screen the optimal preparation process, which were assessed by recombination rate. In the study, four factors were explored, which were the drug concentration, reaction time, reaction temperature and reaction ratio. At the same time, UV, IR and microscope analysis technique were used to analyze the structure of phospholipid complex. Results The optimal preparation conditions of florfenicol phospholipid complex were as follows: the reaction solvent was tetrahydrofuran, the quality ratio of florfenicol and phospholipid was 1:2, the reaction temperature was 45 ℃, and the reaction time was 2h. When doing as the optimal preparation conditions, the highest recombination rate was 95%. Conclusion The reaction ratio, the concentration offlorfenicol and the reaction time had geater impact on the formation of phospholipid complex, and the reaction temperature also had some impact. Chemical changes did not occur in the formation process, but the phospholipid complex was essentially different with the simple physical mixtures.
