摘 要:目的:优选石韦总黄酮的大孔树脂纯化工艺条件,为该成分的制剂开发提供参考。方法:采用紫外-可见分光光度法测定总黄酮含量,检测波长510 nm。以总黄酮含量为指标,通过静态吸附-洗脱试验筛选大孔树脂型号,利用单因素试验考察上样量、径高比、洗脱剂用量等因素对石韦总黄酮大孔树脂纯化工艺的影响。结果:D101型大孔树脂吸附-洗脱性能最好,最佳纯化工艺条件为树脂柱径高比1∶10-1∶8,树脂-生药(1∶2.73),上样液质量浓度0.5 g·m L^-1,上样速度2 BV·h^-1,加水7 BV洗脱,加60%乙醇5 BV以流速4 BV·h^-1洗脱。石韦总黄酮纯度达59.06%。结论:D101型大孔树脂对石韦总黄酮具有良好的纯化效果,优选的纯化工艺合理、稳定可行。Objective: To optimize purification process of total flavonoids from Pyrrosiae Folium by macroporous resin. Method: The content of total flavonoids was determined by UV-visible spectrophotometry, detection wavelength was 510 nm. With the content of total flavonoids as index, types of maeroporous resin were screened by static adsorption-elution test, single factor tests were adopted to optimize purification technology with sample volume, ratio of diameter to height, the amount of eluent and so on. Result: D101 macroporous resin showed better adsorptionlution effect than others, optimum purification technology was as following: heightdiameter ratio of 6-10, ratio of resin to crude drug 1: 2.73, the concentration of sample solution 0.5 g ·mL^-1, absorption velocity 2 BV ·h^-1, washed impurity with 8 BV of water, then eluted with 5 BV of 60% ethanol at 4 BV ·h^-1, collected eluent. Under these conditions, purity of total flavonoids was 59.06%. Conclusion: D101 maeroporous resin has good purification effects on total flavonoids from Pyrrosiae Folium, this optimized purification technology is reasonable, stable and feasible.
