摘 要:目的:探讨穿心莲内酯对动脉粥样硬化新西兰兔主动脉微炎症反应、粥样斑块形成及主动脉病理改变的影响。方法:SPF级新西兰家兔36只,雌雄各半,随机分为6组:正常组、模型组、非诺贝特组(25.67 mg·kg-1)、穿心莲内酯高、中、低剂量组(93.34,46.67,23.33 mg·kg-1)。正常组给予普通饲料16周,其余各组一次性耳缘静脉注射牛血清白蛋白,加给予高脂饲料50 g·d-1造模喂养12周,12周后分组继续高脂饲料喂养,各给药组按剂量灌胃给予相应药物,连续给药4周。末次给药后1 h取血清进行血脂水平测定,干扰素-γ(IFN-γ)、白介素-4(IL-4)、白介素-1(IL-1β)及超敏C反应蛋白(hs-CRP)的含量测定,取主动脉检测血管间细胞黏附分子(VCAM-1)、单核细胞趋化蛋白-1(MCP-1)和血小板源性生长因子(PDGF)的mRNA表达以及组织病理学观察。结果:与正常组比较,模型组新西兰兔血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C),IL-1β含量及hs-CRP含量明显升高,高密度脂蛋白胆固醇(HDL-C)含量明显降低,主动脉VCAM-1,MCP-1及PDGF表达明显升高(P<0.01);与模型组相比,穿心莲内酯高、中剂量组新西兰兔血清LDL-C,IFN-γ/IL-4及hs-CRP含量较模型组降低,穿心莲内酯高、中、低剂量组血清TC,TG及IL-1β含量明显降低,HDL-C含量明显升高,主动脉VCAM-1,MCP-1及PDGF表达明显降低(P<0.01)。组织病理学观察穿心莲内酯高剂量组斑块隆起不明显,内皮细胞排列均匀;中剂量组斑块较模型组减少,仍可见部分排列均匀的内皮细胞;低剂量组斑块厚度达动脉全层一半,斑块形成程度较模型组明显减轻。结论:穿心莲内酯抑制炎症的进程,延缓动脉粥样硬化的发展。Objective: To study the effect of andrographolide on aortic micro-inflammation, atheromatous plaque formation and aortic pathology in atherosclerosis rabbit. Method: Thirty six SPF Newzealand rabbits were divided into normal group, model group, fenofibrate group (25.67 mg ·kg^-1 ) , and high, medium and low dose of andrographolide group (93.34, 46.67, 23.33 mg ·kg^-1 ). Except for the normal control group, the rest animals were used to establish atherosclerosis model with bovine serum albumin and high-fat diet for 12 weeks. After the modeling, the rabbits were treated with andrographolide for 4 weeks. Result: Compared with normal group, the level of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL- C), interleukin-1β (IL-1β) and high-sensitivity C-reactive protein (hs-CRP), the expression of vascular cell adhesion molecule-1 (VCAM-1), monocyte chemoattractant protein-1 (MCP-1) and platelet derived growth factor (PDGF) of the model group were obviously increased, the level of high density lipoprotein cholesterol (HDL-C) was reduced, the difference was statistically significant (P 〈 0.01 ). Compared with model group, the ratio of Interferon γ/Interleukin 4 (IFN-γ/IL-4), the level of TC, TG, LDL-C, IL-lfl and hs-CRP, the expression of VCAM-1, MCP-1 and PDGF mRNA of the andrographolide group were obviously reduced, the level of HDL-C was increaed, the difference was statistically significant (P 〈 0.01 ). Conclusion: Andrographolide could inhibit the inflammation process, and delay the development of atherosclerosis.
