一株致病性血清2型猪链球菌的多位点序列分型及其灭活后对小鼠的免疫保护效果评价

摘　　要：为鉴定一株2012年分离自广东省的致病菌并评价其免疫原性,本研究对该分离株进行分型鉴定和MSLT分型分析。鉴定结果显示,该分离株为血清2型猪链球菌(S.suis);其MSLT分型为ST1型,等位基因图谱是1、1、1、1、1、1、1,与高致病性的世界流行株P1/7同型;毒力基因型为Mrp+/Ef+/Sly+,将其命名为GD。进一步使用GD灭活菌以3×108cfu剂量3次免疫小鼠,免疫后分别采用GD,7型、9型S.suis WH和ZD进行攻毒试验。动物试验结果显示,免疫后小鼠抗体水平显著升高;对GD、WH和ZD的保护率分别为87.5%、87.5%和75%;免疫组小鼠血液和脏器内的细菌载量均显著低于对照组,而且无明显病变。本研究结果表明,GD灭活疫苗可以对不同血清型S.suis攻毒提供有效的免疫保护,可以作为预防S.suis病的有效灭活疫苗候选株。A pathogenic Streptococcus suis isolated from Guangdong province in 2012 was identified by multilocus sequence typing （MSLT） and the immunogenicity was evaluated in mice. The results showed that the isolate was S.suis serotype 2; and MLST tests was ST1, allelic spectrum was 1, 1, 1, 1, 1, 1, 1, which was highly identical to the pandemic pathogenic strain P1/7 with virulence genotype of Mrp＋/EfTSly＋, named GD. The groups of mice were immunized with the inactivated GD at a dose of 3×10^8 cfu for 3 times, and challenged at 7 days post the bosting with S.suis 2 GD, S.suis 7 WH, and S.suis 9 ZD, respectively. Animal experiments showed that the antibody levels in immunized mouse gruops were significantly higher than that of control groups, and the protection rates were 87.5%, 87.5% and 75% respectively against GD, WH and ZD strain. Bacteria levels in blood and organs of mice in immunized group were significantly less than the control group, and no significant pathological changes were observed. These results showed that the inactivated S.suis GD was an premising vaccine candidate to provide effectiveimmune protection against homologous and heterologous S.suis.
