摘 要:目的:观察丝裂霉素(MMC)联合雷帕霉素(RAPA)对体外培养的人膀胱癌T24细胞抑制作用是否比单药大。方法:体外培养人膀胱癌T24细胞,通过MTT实验及相关公式计算,得出MMC和RAPA的IC50值,以此值及相关药量应用于膀胱癌T24细胞,经24、48h,比较两种药物单用及联合用药(全量及半量)对癌细胞的抑制率,得出抑制率最高的药物(两种单药或联合用药)。采用方差分析、SNK-q检验分析得出结果。结果:MMC、RAPA的IC50值分别为12μg/ml、125μg/L(最后实验所用)。两种药物单用及联合用药的24h抑制率最高的是MMC联合RAPA,全量及半量联合的抑制率差异无统计学意义,两者抑制率分别为63.64%及73.68%;MMC联合RAPA比单药的抑制率高,差异有统计学意义;与单药48h抑制率比较,MMC联合RAPA的抑制率差异无统计学意义(其中MMC联合RAPA全量联合的抑制率最大,为98.53%),但是高于RAPA的抑制率,差异有统计学意义。结论:MMC联合RAPA对体外培养的人膀胱癌T24细胞抑制作用比单药大。Objective: To compare the inhibition effect between single use of mitomycin C (MMC) or rapamycin (RAPA) and drug combination on bladder cancer T24 cell line in vitro. Method: By cells culturing in vitro, MTT and some formulas were used to obtain the IC50 of the two drugs. T24 cells were incubated with the IC50 of the two drugs alone and in drug combination (all and half of IC50 ) to obtain the largest inhibition ratio. The experiment was timed two periods that 24 and 48 hours (after treated with drugs for g4 hours and 48 hours). SPSS was used in statistical analysis with analysis of variance and SNK-q test. Result: The IC50) of MMC, RAPA were 12μg/ml, 125 μg/ml respectively (the dose of the experiment). The largest inhibition ratio of every group of two drugs and drug combination was the drug combination of MMC and RAPA (half and all of IC50 ) whose inhibition ratio of 24 hours were 73. 68% 63.64%. The inhibition ratio of drug combination was larger than single use of MMC, RAPA and the difference was statistical significance. The largest inhibition ratio in two drugs and the drug combination was MMC, the combination of MMC and RAPA (all of IC50 98. 53%) of 48 hours, which the difference was not statistical significance. The lowest inhibition ratio in all drugs and the drug combination was RAPA, which the difference showed statistical significance. Conclusion: The drug combination of MMC and RAPA shows a larger inhibition ratio on bladder cancer T24 cell line in vitro than MMC or RAPA used alone.
