摘　　要：目的探讨天然雌激素及植物雌激素对香烟烟雾提取物(cigarette smoke extract,CSE)导致的人皮肤弹性组织变性的影响。方法光镜下观察CSE染毒及加入不同雌激素干预后成纤维细胞(fibroblast,FB)的形态变化;MTT比色法测定不同浓度、不同种类的雌激素对CSE干预的FB增殖活性的影响;荧光定量PCR法检测FB中Fibulin-3,Fibulin-4,Fibulin-5,Elastin在CSE及雌激素干预后的表达变化。结果光镜下可见CSE染毒后的FB失去正常形态,加入雌激素干预后形态趋于正常;MTT结果显示,CSE对体外培养的FB有抑制作用,加入17β-雌二醇、大豆异黄酮、葛根素均可使其增殖率有不同程度的升高;荧光定量PCR结果显示,CSE处理后Fibulin-3,Fibulin-4,Fibulin-5,Elastin mRNA表达降低,加入17β-雌二醇、大豆异黄酮、葛根素干预后mRNA表达有不同程度的回升(P<0.05)。结论 17β-雌二醇、大豆异黄酮、葛根素对香烟烟雾提取物诱导的人皮肤弹性组织变性有不同程度的逆转性保护作用。Objective To explore the natural estrogens and phytoestrogens impact on cigarette smoking caused elastosis in human fibrobllast. Methods The morphological changes of fibroblasts after 24 hours＇ treatment with ciga- rette smoke extract, natural estrogens and phytoestrogens were observed with invert microscope. The inhibitory effect with CSE and estrogens on fibroblast were determined by the MTT test. The mRNA expressions of Fibu- lin-3,-4,-5 and elastin were detected by fluorescence quantitative PCR assay. Results Light microscopy showed that fibroblasts lost its normal morphology after exposed to CSE and tends to normal after added estro- gens. MTF assay showed a significant inhibition of fibroblasts cultured with CSE in vitro. Its proliferation rate rises when 17β-estradiol, soy isoflavones or puerarin was added to the fibroblasts after cigarette exposure. The results of RT-PCR showed that Fibulin-3,-4,-5 and elastin expression level were significantly decreased after the treatment with CSE and their expressions were upregulated after adding estrogens （ P 〈 0.05 ）. Conclusion 17β-estradiol, soy isoflavones,puerarin has protective effect on cigarette smoke extract caused elastosis in human skin fibroblasts.
