摘　　要：目的研究同型半胱氨酸(Hcy)对人脐静脉内皮细胞(HUVEC)运动能力以及肌球蛋白轻链激酶(MLCK)表达的影响。方法不同浓度的Hcy处理人脐静脉内皮细胞后,MTT法检测Hcy对HUVEC增殖的影响,划痕实验检测Hcy对细胞迁移的影响,免疫组化法及Western blot法检测MLCK表达的变化。结果随着浓度的升高,Hcy对HUVEC的增殖有显著的抑制作用,对HUVEC的迁移有明显的促进作用,免疫组化法及Western blot法结果显示Hcy可以增强MLCK的表达。结论 Hcy对HUVEC的迁移具有促进作用,可能是Hcy通过上调MLCK的表达所致。Objective To explore the effect of homocysteine（Hcy） on movement ability and expression of myosin light chain kinase（MLCK） in human umbilicalvein endothelial cells（HUVEC） . Methods MTT assay was detec-ted to measure the proliferation of HUVEC treated with different concentrations of homocysteine. The effect of Hcy on migration of HUVEC was measured by wound healing assay. The expression of MLCK was analyzed by immuno-histochemistry and Western blot. Results With the increase of concentration,Hcy could inhibit the proliferation of HUVEC and promote the migration of HUVEC remarkably. Immunohistochemistry and Western blot showed that Hcy could enhance the expression of MLCK significantly. Conclusion Hcy can promote the migration of HUVEC, which may relate to the up-regulation of expression of MLCK.
