摘 要:目的探讨新生7d大鼠接受不同浓度七氟醚麻醉后对其性成熟期(出生后55d~60d)空间学习记忆能力的影响。方法出生7d龄sD大鼠48只,体重12g~16g,雌雄各半,采用随机数字表法分为3组(每组16只):3%七氟醚吸入6h组(A组)、1.5%七氟醚吸入6h组(B组)和空白对照组(C组)。新生大鼠接受麻醉后次日(8d)每组各取8只,采用直接断头法处死,取大脑海马CAl区组织,利用比色法测定乙酰胆碱酯酶(acetylcholine esterasc,AchE)及胆碱乙酰转移酶(cholineacetyhr ansferase,ChAT)的活性。剩余大鼠继续生长至性成熟期(55d~60d)时,先利用Morris水迷宫实验测试其空间学习记忆能力,实验结束后再测定大脑海马CA1区AchE及ChAT的活性。结果①8d时:A组及B组AchE活性分别为(0.34±0.07)U/mg和(0.33±0.07)U/mg,均较C组增加(P〈0.01);A组及B组ChAT活性分别为(81±22)U/mg和(96±29)U/mg,均较C组降低(P〈0.01)。②性成熟期时:Morris水迷宫实验中,A组和B组平均逃避潜伏期、原平台象限活动时间及穿越原平台次数与c组比较,差异均无统计学意义(侈0.05);60d时,AchE活性、ChAT活性各组间差异均无统计学意义(P〉0.05)。结论新生7dSD大鼠接受不同浓度七氟醚麻醉后可短暂性升高大脑海马CAl区AchE活性,降低ChAT活性表达,但不影响其发育至性成熟期时的空间学习记忆能力。Objective To investigate the effect of different concentrations sevoflurane on spatial learning and memory functions into sexual maturity period (55 d-60 d) in neonatal rats (7 d). Methods Forty-eight SD rats of both sexes aged 7 days weighing 12 g-16 g were randomly divided into 3 groups(n=16): group A and B inhaled 3% and 1.5% sevoflurane in oxygen( 1 lJmin) for 6 h respectively, control group(group C)[inhaled oxygen( 1 L/min) only]. The effect of sevoflurane anesthesia on spatial learning and memory functions were assessed using Morris water-maze test in sexual maturity period (55 d-60 d). The rats were killed immediately on the next day after inhalation anesthesia (8 d) and after the last Morris water-maze test (60 d) respectively, and the hippocampal CA1 neurons were removed for the expression of acetylcholine esterase (AchE) activity and choline acetyltransferase (CHAT) activity using Colorimetry. Results (1) On the next day of inhalhtion (8 d) : the AchE activity expression of group A and group B were (0.34±0.07) U·mg-1·prot-1 and (0.33±0.07) U·mg-1·prot-1, both up-regulated over the group C (P〈0.01), the ChAT activity expression of group A and group B were (81±22) U·mg-1·prot-1and (96±29) U·mg-1·prot-1], both de-regulated over the group C (P〈0.01). (2) In sexual maturity period, there were no significant differences in escape latency, explore time in the inherent platform quadrant, number of cross platform and AchE activity, ChAT activity between experimental groups and control group (P〉 0.05). Conclusions Different concentrations sevoflurane anesthesia in neonatal rats can transiently increase the expression of AchE activity and reduce the expression of ChAT activity in hippocampal CA1 neurons,but does not affect the rats'spatial learning and memory functions in sexual maturity period.
