摘  要:目的:研究鱼油对母鼠以及新生仔鼠海马组织中ASIC1a、ASIC1b、ASIC2a、ASIC2b基因表达的影响。方法:将32只雌雄各半的成年SD大鼠随机分为阴性组与鱼油组,每组16只,雌雄各半。雌鼠受孕后开始饲喂鱼油,并连续饲养60 d,之后采用实时荧光定量PCR技术检测母鼠与仔鼠海马组织中ASIC1a、ASIC1b、ASIC2a、ASIC2b m RNA的表达。结果:与阴性组相比,鱼油组母鼠和仔鼠海马组织中ASIC1a和ASIC1b m RNA的相对表达量显著降低;ASIC2a与ASIC2b m RNA的相对表达量显著增高。结论:孕期添加适当剂量的鱼油,能够降低母鼠和新生仔鼠海马组织中ASIC1a和ASIC1b的基因表达,增加ASIC2a与ASIC2b的表达。Purpose: To investigate the effect of fish oil on gene expression of ASIC1 a, ASIC1 b, ASIC2 a, ASIC2 b in hippocampus of maternal rats and neonatal rats. Methods: A total of 32 adult SD rats with half males and half females were randomly divided into control group and fish oil group. There were 16 rats with half females and half males in each groups. when the female rats were pregnant, they were continuously fed with fish oil for 60 days. After that, Real-Time PCR was used to test the m RNA expression of ASIC1 a, ASIC1 b, ASIC2 a, ASIC2 b in hippocampus of maternal rats and offspring rats. Results: Compared with the control group, the relative m RNA expression of ASIC1 a and ASIC1 b in maternal rats and offspring rats' hippocampus of fish oil group significantly declined(P〈0.05); the relative m RNA expression of ASIC2 a and ASIC2 b in maternal rats and offspring rats' hippocampus of fish oil group obviously increased(P〈0.05). Conclusions: the optimal dosage of fish oil supplementation during the perinatal period could reduce the expression of ASIC1 a and ASIC1 b, increase the expression of ASIC2 a and ASIC2 b in hippocampus of maternal rats and offspring rats.
