摘 要:钙蛋白酶抑制蛋白(CAST)基因是影响肉质性状的重要的候选基因。研究选取40只巴美肉羊为试验对象。采用相对定量PCR技术对不同月龄巴美肉羊背最长肌、臂三头肌、股二头肌中CAST基因的表达规律及其与肉质的相关性进行研究。分析基因表达量与肉质指标的相关性。实验结果表明:3个测试部位中,CAST基因的表达规律为臂三头肌(Q)>股二头肌(H)>背最长肌(B)。在背最长肌中,CAST基因表达量与剪切力(r=0.619)、红度值(r=0.719)、黄度值(r=0.222)、亮度值(r=0.438)、眼肌面积(r=0.382)呈正相关,与p H1(r=-0.643)、p H2(r=-0.330)呈负相关。在臂三头肌中,CAST基因表达量与剪切力(r=-0.140)、红度值(r=-0.023)、p H1(r=-0.684)呈负相关,与黄度值(r=0.263)、亮度值(r=0.614)、p H2(r=0.121)呈正相关。在股二头肌中,CAST基因表达量与剪切力呈显著正相关,相关系数0.989(p=0.011);与红度值(r=0.928)、黄度值(r=0.213)、p H2(r=0.734)呈正相关;与亮度值(r=-0.264)、p H1(r=-0.812)呈负相关。Calpastatin(CAST) gene was important candidate gene for meat quality. Correlation of the expression with meat quality had been investigated. Relative quantitation PCR was used to investigate the expression of CAST in 40 Bamei sheep of different month ages. The shear force, p H, color were investigated after slaughter. The correlation between the expression and meat quality traits were analyzed. Results showed that the expression pattern of CAST gene was triceps biceps femorislongissimus dorsi. The expression of CAST gene in longissimus dorsi had positive correlation with shear force(r=0.619), red/green color(r=0.719), yellowness(r=0.222), lightness(r=0.438), loin area(r=0.382) and had negative correlation with p H1(r=-0.643), p H2(r=-0.330). The expression of CAST in triceps had negative correlation with shear force(r=-0.140), red/green colors(r=-0.023), p H1(r=-0.684) and had positive correlation with yellowness(r=0.263), lightness(r=0.614), p H2(r=0.121). The expression of CAST in biceps femoris had significant positive correlation with shear force(r=0.989, p=0.011), and had positive correlation with red/green colors(r=0.928), yellowness(r=0.213), p H2(r=0.734). The expression of CAST was negative correlation with lightness(r=-0.264) and p H1(r=-0.812).
