摘  要:为了提高热水法浸提猴头菇多糖得率,应用单因素试验和响应面法对热水法浸提猴头菇多糖工艺进行优化。用单因素试验对浸提工艺进行初步优化,结果表明水料比、浸提温度和浸提时间对多糖得率均有显著影响。在此基础上,用响应面法优化,构建多糖得率的三元二次回归方程。通过对响应面图和等高线图进行分析得到回归拟合方程的决定性系数(R2Adj)为0.9825,表明响应面模型能够代替试验真实点对多糖得率进行预测。响应面优化结果表明,热水法浸提猴头菇多糖的最优工艺为:水料比33 m L/g、浸提温度92℃、浸提时间134 min。在最优工艺条件下,多糖得率为4.98%,非常接近响应面预测值,说明采用响应面法优化得到的浸提工艺参数可靠。In order to improve the yield of polysaccharide, hot water extraction process of polysaccharide from Hericium erinaceus was optimized by mono-factorial experiment and response surface methodology(RMS). By the mono-factorial experiment, the results showed that the significant external factors affecting yield of polysaccharide were the ratio of water to material, extraction temperature and extraction time. Based on these results, RSM was employed in the present work and a quadratic equation for yield of polysaccharide was built. By analyzing the response surface plots and their corresponding contour plots as well as solving the quadratic equation, the experimental values were shown to be significantly in good agreement with predicted values since the adjusted determination coefficient was 0.9825. The optimum process parameters for hot water extraction process were obtained as: particle size 30, the ratio of water to material 33 m L/g, extraction temperature 92 ℃, extraction time 134 min, 4.98% of the yield of polysaccharide was achieved. The predicted values for optimization process conditions were in good agreement with experimental data, indicating that the optimum process parameters by RMS were reliable.
