摘  要:目的本文旨在优化十子代平方中中药复方多糖的提取工艺,测定其含量并分离纯化得精制的总多糖。方法采用正交实验法优化十子代平方总多糖的提取工艺,用苯酚-硫酸法测定十子代平方总多糖的含量,通过脱蛋白,醇沉,透析等分离纯化十子代平方总多糖。结果十子代平方总多糖的提取最佳条件为70℃,超声提取60min,超声功率为40%,提取次数为3次,料液比为1∶20。经过纯化,总多糖的含量逐步提高,去蛋白后的多糖含量为43.93%,醇沉提高到52.97%,经过透析提高到63.19%。结论本实验为中药复方多糖的分离纯化条件提供研究基础。Objective To optimize the extraction process of compound polysaccharides in ten offspring square, and to determine its content and separate and purified polysaccharide. Methods Polysaccharide extraction in ten offspring square was optimized by orthogonal experiment method, the total polysaccharide content was determined by phenolsulfuric acid method through the removal of protein, ethanol precipitation, dialysis purification. Results The optimum extraction conditions of polysaccharide in cross square was 70 ℃, ultrasonic 60min, ultrasonic power 40%, extract 3 times, solid-liquid ratio was 1.20. After purification, gradually increase the content of polysaccharide and protein- polysaccharide content of 43. 93%, after alcohol up to 52. 97%, after dialysis up to 63.19%. Conclusion This study provides for the isolation and purification of polysaccharides of complex conditions are established.
