摘 要:目的了解HPA-1~6、15系统基因多态性在广西儿童慢性特发性血小板减少性紫癜(ITP)分布情况,探讨儿童慢性ITP与HPA-1~6、15系统等位基因的相关性。方法采用聚合酶链反应技术(PCR)结合直接测序方法,对儿童慢性ITP患者46例(ITP组)和正常健康儿童48例(对照组)进行HPA-1~6、15共7个抗原系统基因分型。结果 ITP组HPA-2、15系统基因频率明显异于对照组(P〈0.05),其中HPA-2a及HPA-15a基因频率低于对照组,HPA-2b及HPA-15b基因频率高于对照组,其余系统等位基因频率在2组间的未发现有明显差别(P〉0.05)。结论HPA-2、15抗原系统多态性可能与儿童慢性ITP的易感性有关。Objectives To study the distribution of genetic polymorphisms of human platelet antigen HPA-1 - 6,15 sys- tems among Guangxi pediatric patients with chronic idiopathic thrombocytopenic purpura(CITP) and to investigate the rela- tionship between CITP and the genetic polymorphism of HPA-1 - 6,15 systems. Methods The genotypes of HPA-1 - 6 and 15 in the ITP group (n = 46) and the healthy control group (n = 48) were identified using polymerase chain reaction (PCR) combined with direct sequencing. Results The allele frequencies of HPA-2, 15 were significantly different between the ITP group and the control group (P =0. 004). The frequency of HPA-2a, 15a in ITP group was lower than the control group. The frequency of HPA-2b, 15b was higher than the control group. There were no significant statistical differences between the frequencies of other alleles ( P 〉 0. 05 ). Conclusion This study shows that genetic polymorphism in HPA-2, 15 antigen system may increase susceptibility to chronic ITP in children.
