摘 要:利用均匀分布于棉花26条染色体上的39对SSR核心引物对16个不同来源的棉花品种样品进行区别与鉴定实验,探讨了棉花品种鉴定的取样方案和位点异同判读方法。通过比较不同取样量样品包含的遗传信息,确定了基本的取样量,并将样品间位点异同类型分为相同位点、疑似相同位点和差异位点。据此对16个品种进行位点异同性分析,结果表明同一品种的不同样品间差异位点数均在0~2间,而不同品种间的差异位点数介于5~25。结合品种间差异位点的阈值分析,研究认为当两样品间差异位点数〉2时,即判定为不同品种;当差异位点数≤2时,即判定为近似或极近似品种。To assess the sampling mode and site determination methods for identifying cotton varieties, 39 pairs of SSR primers distributed evenly on the 26 chromosomes of cotton were screened in 16 cotton varieties, and showed high polymorphism and good repeatability. The basic sampling amount was determined by comparing the genetic information of different sampling modes. The sites were classified into three types: the same locus, the suspected same locus, and the discrepant locus, according to the similarities and differences of sites. The similarities and differences of sites in the 16 varieties were analyzed using the above scheme. The results showed that the number of discrepant loci for the same variety was between 0 and 2, and the number for different varieties ranged fi'om 5 to 25. Based on the analysis of the discrepant locus threshold between different varieties, samples can be determined to be different when the number of discrepant loci is more than 2, and similar when there are fewer than 2 discrepant loci.
