摘 要:通过陆地棉与海岛棉杂交并与陆地棉回交,获得1个染色体片段代换系苏VR043,抗江苏非落叶型黄萎病菌系BP2。分子检测表明,苏VR043兼有陆地棉苏棉8号的遗传背景和海7124的D4染色体片段。以苏VR043和苏棉8号为亲本构建包含176个单株的F2群体,通过标记分析和F2:3家系抗病鉴定,发现抗病性状与标记NAU3392连锁,p值为2.3×10-12。根据棉花D组染色体测序结果,参照置换区段的基因组序列,合成92对SSR引物;PCR扩增分析发现,有5对引物在苏棉8号和苏VR043之间表现多态性,并将抗病主效QTL定位在标记ZHX32和NAU3392之间,标记间遗传距离为3.2 c M,QTL解释表型变异64.8%。同时参照棉花D组测序结果,提取对应的置换区段序列,预测包含63个基因,基因聚类分析表明7个基因参与抗逆反应,其中1个基因与抗病相关。To dissect the Verticillium wilt resistance QTL and find molecular markers closely linked with the resistance trait,we created a new material.By crossing Gossypium hirsutum L.cv.Sumian 8 as the recipient parent and Gossypium barbadense L.cv.Hai 7124 as the donor parent,we generated a chromosome segment introgression line (CSIL),SuVR043,with higher resistance to non-defoliating BP2 isolates of Verticillium dahliae.SuVRo43 was characterized as carrying one homozygous chromosome 22 (D4) segment represented by the SSR marker NAU3392 from G.barbadense cv.Hai 7124 in the background of the upland cotton Sumian 8.We constructed an F2 mapping population,SuVR043 × Sumian 8,containing 176 individuals.Based on SSR markers and F2:3 phenotypes for resistance to Verticillium wilt strain BP2,single marker analysis showed the resistance phenotype was linked to the maker NAU3392 with a p value of 2.3×10^-12.Using the recently released cotton D genome sequence information as a reference,we extracted the introgression segment sequence and designed 92 SSR primer pairs,from which five polymorphic SSR loci were detected between SuVR043 and Sumian 8 and used to map the Veaicillium dahliae resistance QTL on the introgression segment.The QTL was anchored between the ZHX32 and NAU3392 loci with a distance of 3.2 cM,and explained 64.8% of the phenotypic variation.Moreover,we predicted 63 candidate genes using the cotton D genome sequence information.The results of Gene Ontology analysis indicated that these genes were involved in different biological processes,with seven of them participating in the stress signal transduction and one participating in biotic stress responses.
