摘 要:目的:在视网膜水平细胞上研究激活NMDA受体对GABA转运体的调节作用。方法:采用膜片钳全细胞记录,分析水平细胞NMDA受体在激活前后的GABA转运体电流变化,并分析Ca2+在其中的作用。结果:当灌流0.1mmol/L的NMDA 30s后,1 mmol/L GABA所引发的GABA转运体电流下降到了对照水平的(78.01±3.10)%(n=5);当细胞外Ringer’s液的Ca2+被去除后,NMDA的抑制效应消失;当胞内Ca2+库的Ca2+释放和重吸收被阻断,或者胞内Ca2+被BAPTA缓冲后,NMDA的抑制效应减弱。结论:激活视网膜水平细胞的NMDA受体会抑制GABA转运体电流,细胞内Ca2+参与调节过程。Objective: The modulation of NMDA receptors on GABA transporters was investigated on retinal horizontal cell. Methods: Changes of GABA transporter current were observed after activation of NMDA receptors via whole cell recording,and intracellular Ca^2 + was also studied. Results: GABA transporter currents( by 1 mmol / L GABA) were decreased to 78.01% ± 3.10%( n = 5) of control level after application of 0.1 m M NMDA. The inhibition of GABA transporters was attenuated by blockage of Ca^2 + release and uptake of intracellular Ca^2 + store. The effect was also decreased when BAPTA was added to intracellular solution. Conclusion: inhibition of GABA transporters can be modulated by NMDA receptors on retinal horizontal cell,and intracellular Ca^2 + plays an important role.
