摘 要:以黄根为试材,采用单因素试验和正交试验相结合的方法,考察超声波频率、超声波功率、乙醇浓度、提取时间、固液比、提取温度对黄根总黄酮提取率的影响,以优化超声波提取黄根中总黄酮的工艺条件。结果表明,最佳提取工艺:超声波频率为高频(61 KHz),超声波功率为60 W,乙醇浓度100%,提取时间105 min,固液比1:15,提取温度65℃。另外,同一产地黄根植株不同部位总黄酮含量高低顺序大致为,结果植株:根>叶>茎>侧枝;不结果植株:叶>根>茎>侧枝。试验结果表明优选出的提取工艺稳定、重复性好,为黄根中总黄酮成分的提取提供了试验依据。Taking Prismatomeri tetrandra as material, using the method of single factor test and orthogonal test, the effect of ultrasonic frequency, ultrasonic power, alcohol concentration, extracting time, ratio of solid and liquid on extracting rate and extracting temperature of flavonoid from Prismatomeri tetrandra were studied to optimize extracting technique. The results showed that the optimal extracting conditions were ultrasonic frequency 61 kHz, ultrasonic power 60 W, alcohol concentration 100%, extracting time105 min, the ration of solid and liquid 1:15 g/mL, extracting temperature 65℃. On the other hand, quantitative order of flavonoid in different sections of Prismatomeri tetrandra from the same area was, results plants: root〉 leaf〉 stem〉 collateral; fruitless plants: leaf〉 root〉 stem〉 collateral. The stability and repeatability of this technique were good. It could provide the basis for extracting flavonoid from Prismatomeri tetrandra.
