多西紫杉醇在肝癌细胞裸鼠肝脏移植瘤中对P53、VEGF作用的研究

摘　　要：目的探讨多西紫杉醇在肝癌细胞株SMMC-7721裸鼠皮下移植瘤中与血管内皮生长因子(VEGF)、肿瘤抑制蛋白(p53)表达的研究.方法建立肝癌细胞株SMMC-7721裸鼠皮下移植瘤模型,建模成功后,将20只裸鼠分成两组,多西紫杉醇组及肿瘤对照组.3周后处理裸鼠,应用免疫组化Envision法检测实验组与对照组肝癌组织中p53、VEGF表达情况.结果实验组肝癌组织中p53表达阳性率41.26%,显著低于对照组肝癌组织的p53表达阳性率87.16%,两者间差异有统计学意义(P<0.05).实验组肝癌组织中VEGF表达阳性率为36.79%,显著低于对照组肝癌组织的VEGF表达阳性率68.27%,两者间差异有统计学意义(P<0.05).结论p53在诱导肝癌细胞凋亡中起到重要作用,VEGF过度表达在肝癌微血管生成过程中起重要作用,多西紫杉醇可降低p53、VEGF的表达,从而起到移植肿瘤生长及转移的作用.Objective To observe the effect of docetaxel on the expression of p53 and VEGF in hepatoma cells SMMC-7721 transplanted tumor model in nude mice. Methods Twenty nude mice with transplant subcutaneous were randomly divided into 2 groups （n = 10） , docetaxel group and control group. The expression of p53 and VEGF were compared between two groups using envision immunohistochemistry. Results After 3 weeks with treatment, p53 in control group was 87.16%, while in docetaxel group was 41.26%. Obviously, the expression of p53 in docetaxel group was lower than that in control group （P 〈 0.05） . VEGF in docetaxel group was 36.79 %, which was lower than that in control group （68.27%） , and the difference was statistically significant （P 〈 0.05） Conclusion Docetaxel can inhibit the growth, invasion and metastasis of hepatoma cells SMMC-7721 by lowering the expression of p53 and VEGF.
