摘 要:目的探讨颅脑损伤(traumatic brain injury,TBI)大鼠皮质脑组织缺氧诱导因子-1α(HIF-1α)和葡萄糖转运蛋白-3(GLUT-3)的表达变化,并进行相关性研究。方法 108只SD大鼠随机分为正常对照组(n=12)和实验组(n=96)。实验组采用自由落体打击法建立大鼠颅脑损伤模型,正常对照组不做处理。分别采用RT-PCR及免疫组化检测伤后1、4、8、12 h及1、3、7、14 d挫伤灶周围HIF-1α和GLUT-3 m RNA及蛋白的表达。结果 RT-PCR结果显示:实验组HIF-1α和GLUT-3 m RNA在伤后4 h升高,12 h达高峰,7 d基本正常;伤后4 h^3 d与正常对照组比较,差异均有统计学意义(P<0.05)。Western blot检测显示:实验组HIF-1α和GLUT-3蛋白在伤后4 h表达升高,1 d达高峰,7 d降至正常水平;伤后4 h^3 d与正常对照组差异均有统计学意义(P<0.05)。相关性分析表明:HIF-1α与GLUT-3在m RNA及蛋白水平均呈正相关(r=0.894,P<0.05;r=0.921,P<0.05)。结论 TBI后脑组织HIF-1αm RNA及蛋白的表达上调,可能介导GLUT-3的表达增加,从而发挥神经保护作用。Objective To investigate the expression of hypoxia inducing factor 1 alpha(HIF-1α) and glucose transporters-3(GLUT-3) in the cerebral cortex in rats with traumatic brain injury and analyze the correlation between them.Methods One hundred and eight SD rats were randomly assigned to normal control group(n = 12) and experimental group(n = 96).The TBI model was established in experimental group by the freely falling body injury method and no injury was performed in normal control group.Expressions of HIF-1α and GLUT-3 m RNA and protein were detected in the contusion area by RT-PCR and Western blot 1,4,8,12 h and 1,3,7,14 d after injury.Results RT-PCR results showed that the expressions of HIF-1α and GLUT-3 m RNA in experimental group increased at 4 h,reached peak at 12 h and decreased to normal 7 d after injury.Compared with normal control group,the expressions of HIF-1α and GLUT-3 m RNA were significantly different during the period from 4 h to 3 d after injury in experimental group(P〈0.05).Western blot results showed that the expression of HIF-1α and GLUT-3 protein in experimental group increased at 4 h,reached peak at 1 d and decreased to normal at 7 d after injury.Compared with normal control group,the expressions of HIF-1α and GLUT-3 protein were significantly different during the period from 4 h to 3 d after injury in experimental group(P〈0.05).There were positive correlations between HIF-1α and GLUT-3 in m RNA and protein expressions(r = 0.894,P〈0.05;r = 0.921,P〈0.05).Conclusion The expression of HIF-1α m RNA and protein in the brain cortex increases after TBI,which may mediate increase of GLUT-3 expression,thus playing a role in neuroprotection.
