摘  要:消化酶在营养生理中具有重要作用,试验首先对海蜇不同组织中主要消化酶活力进行研究,然后研究了不同温度和pH下海蜇丝状附属器、胃丝的蛋白酶、淀粉酶活性。分别采用福林-酚法、淀粉-碘比色法、聚乙烯橄榄油乳化液水解法和3,5-二硝基水杨酸法测定蛋白酶、淀粉酶、脂肪酶和纤维素酶活力。结果表明,仅在海蜇的丝状附属器(Filiform appendage,FA)和胃丝(Gastric cirrum,GC)能够检出明显的蛋白酶和淀粉酶活力,而无脂肪酶和纤维素酶活力。随着温度的升高,各种酶的活性在一定范围内均表现为升高趋势。在不同组织中同一种酶活性存在差异。两类能够检出酶活性的组织中,淀粉酶的最适温度均为27℃,蛋白酶的最适温度不同,丝状附属器为52~62℃,胃丝为42℃。pH对海蜇的丝状附属器和胃丝的消化酶活力具有显著影响(P〈0.05)。2种组织中,淀粉酶的最适pH均为7,蛋白酶活力随着pH的升高出现两个相对稳定的峰值,丝状附属器为3和8,胃丝为3和9。Digestive enzymes play a very important role in the nutrition physiology. First of all, a variety of primary di- gestive enzyme activities on jellyfish were studied, then studied the protease and amylase activities in filiform appendage and gastric cirrum with different temperature and pH. To measure the activity of protease, amylase, lipase and cellulase , the Folin--phenol reagent method, the starch--iodine spectrophotometric method, the polyethylene olive oil emulsion hydrolysis method and the 3,5- 2 nitro salicylic acid method, were used respectively. The results showed that : only the protease and amylase activities in filiform appendage (FA) and gastric cirrum (GC) could be detected. There weren't lipase & cellulose activities in sorts of tissues. The digestive enzymes activities increased with the increasing of temperature at a given range. The digestive enzyme activity in different tissues was different as well. The optimum temperature for amylase in two kinds of digestive tissues was 27℃. The optimum temperature for protease in filiform appendage was 52-62℃, and in gastric cirrum was 42℃. The optimum pH for amylase in two kinds of digestive tissues was 7. The highest protease activity of filiform appendage was in two pH ranges: 3 and 8, while the highest protease activity of gastric cirrum was also in two pH ranges: 3 and 9.
