摘 要:目的探讨瓜蒌皮注射液对缺血缺氧的内皮祖细胞(EPC)的保护作用及机制。方法提取大鼠骨髓单个核细胞,M199培养基添加血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(b FGF)诱导培养成EPC。将能吸收凝集素和乙酰化低密度脂蛋白的EPC进行低氧无血清培养以成缺血缺氧损伤模型,同时在培养液中添加系列质量浓度的瓜蒌皮注射液培养上述细胞,MTT法检测其存活率,ELISA法检测细胞VEGF分泌量。结果和对照组相比,缺血缺氧组EPC存活率、VEGF分泌水平均显著低下;瓜蒌皮注射液处理可明显升高细胞存活率、VEGF表达量,作用效果与剂量密切相关。结论瓜蒌皮注射液可能通过促进VEGF的表达,从而提高EPC缺血缺氧条件下的生存能力。AIM To study the protective effect of Trichosanthes pericarpium Injection on hypoxic-ischemic endothelial progenitor cells(EPCs) in vitro.METHODS Mononuclear cells were isolated from rat bone marrow and cultured in M199 medium supplemented with vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(b FGF).Cells that could absorb both lectin and acetylized low-density lipoproteins were selected and co-cultured in low-oxygen and serum-free substrate to form EPCs.Co-culture of EPCs and T.pericarpium Injection was then performed.EPCs cell viabilities were evaluated by MTT and VEGF levels were detected by enzyme-linked immunosorbent assay(ELISA).RESULTS As compared with the treated group,the viabilities and VEGF levels in hypoxic-ischemic EPCs group markedly decreased,while T.pericarpium Injection promoted cell livability and VEGF level of above cells in a dose-dependent manner.CONCLUSION T.pericarpium Injection can obviously enhance the proliferative capacity of hypoxic-ischemic EPCs in vitro by promoting of the expression of VEGF.
