摘 要:目的考察生脉注射液(人参、麦冬、五味子)抗大鼠心肌缺血再灌注损伤的治疗效果,并初步探讨其作用机制。方法采用结扎大鼠冠状动脉左前降支,建立心肌缺血再灌注损伤模型。利用小动物超声心动检测仪检测大鼠射血分数,并同时测定大鼠血液中表征细胞活性、氧化应激、炎症因子等的一系列生化指标。结果结果显示生脉注射液组能明显改善模型大鼠射血分数,抑制心肌缺血再灌注损伤后CK-MB、TNNT2、TNNI3水平上升,降低MDA水平,提高SOD活力,并且抑制TNF-α、IL-1β、IL-6、IL-8等多种炎症因子表达。结论生脉注射液通过多方向药理作用机制,减轻心肌缺血再灌注损伤,发挥心肌保护作用。AIM To study the cardioprotective effects of Shengmai Injection(Ginseng Radix et Rhizoma,Ophiopogonis Radix,Schisandrae chinensis Fructus) against rat myocardial ischemia and reperfusion injury(MIRI) and their mechanisms.METHODS Adult rats subjected to left anterior descending coronary artery(LAD)ligation and release were used for ischemia and reperfusion model.The cardioprotective effects were evaluated by echocardiography and series of biochemical assays related to cell viability,free radicals inflammation.RESULTS The experimental data revealed that Shengmai Injection could obviously improve the left ventricular ejection fraction(LVEF) model rats and relieved the elevation of CK-MB、TNNT2、TNNI3 by MIRI.SMI also reduced MDA and elevated SOD level.Shengmai Injection reduced inflammation related factors including TNF-α,IL-1β,IL-6,IL-8.CONCLUSION Shengmai Injection can exert cardioprotective effects through a polypharmacology mechanism involving alleviating the injury to myocardial ischemia and reperfusion.
