摘 要:目的评价我们研究的血小板冻干技术所获得的冻干血小板的稳定性。方法我们用前期筛选出的,最优血小板预处理液配方技术所获得的冻干血小板,分别置于-20℃、4℃和室温下密封保存,选取0、10、30、90、180 d5个时间点,检测复水化后冻干血小板,以血小板回收率、MPV冻干前后差值、血小板相对聚集率,以及血小板激活后生长因子PDGF-BB、VEGF、TGF-β释放量做为评价指标。结果血小板冻干保存的稳定性研究结果显示:同一时间点,-20℃、4℃和室温3种保存温度之间,血小板回收率、相对聚集率、血小板激活后上清中VEGF和TGF-β含量,均无统计学差异(P>0.05)。结论优选配方预处理后的冻干血小板在室温下保存,与置于4℃和-20℃保存效果相当,冻干血小板室温下可以稳定保存至少6个月。Objective To evaluate the effect of preservation temperature and time on the stability of platelets from freeze-drying. Methods Six bags of optimal pooled platelets were frozen and dried, and then kept under seal at - 20℃, 4℃ and room temperature. Then, the rate of recovery, the difference in platelet volumes before and after freeze-drying, the relative platelet aggregation rate and the amount of activated growth factor PDGF-BB, VEGF, TGF-βafter were measured at 0 d, 10 d, 30 d, 90 d and 180 d. Results There were no significant differences in all indices, including the rate of recover- y, the difference in platelet volumes before and after freeze-drying, the relative platelet aggregation rate and the amount of growth factor ( P 〉 0. 05 ) among - 20 ℃ group, 4 ℃ group and room temperature group at the same time point. Conclusion The frozen and dried platelets prepared and optimized in the laboratory can stay stable for at least 6 months at room temper-ature. The effect of freeze-drying on the platelets was comparable to the results obtained from the 4℃ and -20℃ groups.
