摘 要:目的探讨添加一氧化氮(NO)供体硝普钠(SNP)对储存末期42 d的高原献血者悬浮红细胞和分层红细胞各项指标的影响。方法采自西藏4人份健康献血者血样(400 m L/份),制成悬浮红细胞后保存42 d,将每份悬浮红细胞取出分作2等份,分别归入实验组:使其终浓度为10μmol/L的SNP溶液;对照组:加与SNP等体积的等渗PBS;置于室温下孵育1 h后检测红细胞的变形性,ATP含量、渗透脆性和胞膜上PS、CD47阳性表达率,用Percoll细胞分离液配置不同浓度(1.091、1.0985、1.106、1.113 5、1.121 g/m L)作密度梯度离心后,依次吸取各层红细胞并检测上述指标。结果储存末期高原悬浮红细胞未分层各项指标:胞膜PS阳性表达率,实验组与对照组分别为2.18±0.40 vs 3.05±0.24(P<0.05),其他各项指标2组差异甚小(P>0.05)。经密度梯度离心后,分离出的4层红细胞中,2-4层为可用的红细胞层,PS阳性表达率2、3层细胞,实验组与对照组分别为1.95±0.57 vs 2.3±0.57、2.23±0.39 vs 2.83±0.45(P<0.05),4层细胞2组无明显差异(P>0.05);在100、200剪切率下EI值、胞内ATP含量、渗透脆性Na Cl浓度、胞膜上CD47阳性表达率,3层红细胞中实验组与对照组分别为0.131±0.013 vs 0.110±0.014、0.211±0.018 vs 0.186±0.022、2.753±0.319 vs 2.190±0.234、4.425±0.180 vs 4.801±0.081、52.13±7.00vs49.23±7.44(P<0.05),2、4层红细胞中2组差异很小(P>0.05)。结论添加适当浓度NO供体SNP对储存末期未分层高原献血者浓缩红细胞无明显影响,但对分层3层(中年)红细胞有明显改善作用。Objective To investigate the effect of the addition of sodimn nitroprusside (SNP) supplied by nitric oxide donors (NO) on the indicators of suspended and stratified red blood cells at the end phase of storage period for 42 days drawn from blood donors living at plateau. Methods Blood samples were collected from four healthy male donors living at high altitude (400 mL/bag) , then were eentrifilged to make a suspension of RBC,and lastly were stored at 4℃ for 42 days. The same suspension of RBC was divided into two moieties : an experimental group with addition of SNP of 10 μmol/L con- centration and a control group with addition of equal volumes of SNP and isotonic PBS. After incubation at indoor temperature for one hour, the deformability of RBC, ATP concentration, osmotic fragility, and the rate of positive expression of PS and CD47 were determined. RBCs were separated using Pereoll density gradient centrifuge according to the differential densities (1.091, 1.0985, 1.106, 1.1135, 1. 121g/mL) and removed from onelayer to another. Results At the end phase of stor- age period, the indicators of the red blood cells in suspension that were not separated were as followed : the rates of positive expression of PS on the membrane in the experimental group and control were 2.18± 0. 40 and 3.05±0. 24, respectively (P 〈 0.05 ). Other indicator indices demonstrated no significant differences between two groups. After density gradient eentrifu- gation, there were four layers cells, among which the second, third and fourth layers could be used. The rates of positive ex- pression of PS from both the second and third layers in experimental group and control were 1.95 ± 0. 57 vs 2. 3± O. 57, 2. 23 ± 0. 39 vs 2.83 ± 0. 45, respectively ( P 〈0. 05 ). However, the RBCs from the fourth layer demonstrated no significant sta- tistical difference ( P 〉 0. 05 ).
