摘  要:CLOCK基因是一种与生物节律相关的基因,对维持正常的近日节律起重要作用。本研究旨在分析鸡CLOCK基因在不同组织间的表达以及在同一组织中的节律性表达情况。根据鸡CLOCK基因mRNA序列(登录号:NM_204174)设计特异性引物,利用QRT-PCR方法,检测早上(06:00)、中午(12:00)、傍晚(18:00)3个时间点CLOCK基因在下丘脑、心脏、肝脏、脾脏等17种组织中的表达。结果表明:鸡CLOCK基因在各组织中均有表达,在胸肌中的表达量极显著高于其他各组织(P<0.01),为心脏表达量的1.99倍;在同一种组织不同时间点的表达呈现出一定的节律性,心脏、肝脏、肌胃中表达量持续升高,下丘脑、回肠、空肠、盲肠、腺胃等先升高后降低,且在空肠、盲肠、腺胃中表现出显著的节律性,脂肪中先降低后升高,脾脏、卵巢中持续下降。结果显示,鸡CLOCK基因表达存在时空特异性,并呈现较强的节律性。CLOCK gene plays an important role in maintaining normal circadian rhythms. The purpose of this study was to investigate the mRNA expression level of chicken CLOCK gene in different tissues, and the rhythm of CLOCK gene expression in the same tissue. Specific primers were designed according to the chicken CLOCK mRNA sequence (GenBank ID: NM_204174). Real-time quantitative PCR was used to detect the mRNA expression levels of CLOCK gene in 17 tissues such as hypothalamus, heart, liver, and spleen at the sunrise, noon and sunset 3 time points, re- spectively. The results showed that the CLOCK gene was expressed in all detected tissues. The highest expression level of CLOCK gene was found in breast muscle, which was 1.99-fold compared to that in heart (P〈0.0001). The expres- sion level of CLOCK gene in one tissue showed a certain rhythm in different time points, it could be roughly divided into four groups: continue increasing in heart, liver and gizzard; increasing and then decreasing in hypothalamus, live, ileum, jejunum, cecum, and proventriculus, significant rhythm in jejunum, cecum and proventriculus; decreasing and then increasing in fat; continuing decreasing in spleen and ovary. The mRNA expression level of chicken CLOCK gene shows the temporal and spatial specificity and rhythmicity. The results herein pave the fundamental for functional study of chicken CLOCK gene.
