摘  要:基于核磁共振技术的代谢组学分析奶牛酮病代谢谱,探索奶牛发生酮病时机体代谢的全景式变化,进一步了解奶牛酮病时的代谢物变化规律。收集临床酮病奶牛、亚临床酮病奶牛、健康对照组奶牛共81头,采血后分离血浆并进行预处理,运用NMR技术结合多元统计分析,鉴定差异代谢物,并进行分析讨论。结果表明:获得29个差异代谢物,其中碳水化合物3个、脂类6个、氨基酸14个、其他物质6个。通过查找数据库,发现这些代谢物主要与氨基酸代谢、脂肪代谢和糖类代谢等能量代谢途径相关。代谢组学技术在一定程度上可以揭示奶牛酮病的发生和发展变化,而这些差异代谢物经过进一步验证可能是诊断奶牛酮病的潜在标记物和指标。Metabolic profiling was analyzed by NMR-based metabolomics in order to explore panoramic metabolism changes of ketosis in dairy cows, and to further understand changes of metabolites when ketosis occured. Eighty-one plasma samples of clinical ketosis cows, subclinical ketosis cows and heathy cows were selected. After pretreatment, different metabolites were identified by using NMR technique combined with multivariate statistical analysis, then analyzed and discussed. Twenty-nine metabolites were obtained, in which 3 carbohydrates, 6 lipids, 14 amino acids and 6 other metabolites. Through finding database and analysis of metabolites, these metabolites were mainly related to amino acid metabolism, fat metabolism and carbohydrate metabolism. Metabolomics, to some extent, can reveal change of occurrence and development of dairy cow ketosis. These different metabolites could be potential markers and indicators that diagnosed dairy cow ketosis.
