摘 要:目的:观察补肾活血化痰方对阿尔茨海默病模型大鼠海马细胞外调节蛋白激酶1、蛋白激酶2蛋白表达的影响。方法:建立阿尔茨海默病大鼠模型,造模后第3 d开始用药,补肾活血化痰方配置成1.5g·m L-1的浓度,按12 mg·(kg·d)-1的剂量灌胃给药,盐酸多奈哌齐按0.52mg·(kg·d)-1的剂量灌胃,模型组及假手术组灌胃液为等体积的生理盐水,均每日两次,每天分上午8点和下午4点两次灌胃,共4周,用免疫组化方法观察AD大鼠海马神经元中细胞外调节蛋白激酶的表达变化。结果:模型组大鼠海马CA1区细胞外调节蛋白激酶1、蛋白激酶2阳性细胞的平均光密度明显少于假手术组,且大多细胞的胞浆染色较淡,差异具有显著性(P<0.01);而盐酸多奈哌齐组和补肾活血化痰方组平均光密度较模型组显著增加,可见较多胞浆呈稍浅棕黄色颗粒状的阳性细胞,差异具有显著性(P<0.01);但盐酸多奈哌齐组和补肾活血化痰方与假手术组相比,染色要淡,阳性细胞平均光密度明显减少,具有显性差异(P<0.01);盐酸多奈哌齐组和补肾活血化痰组之间比较,差异不显著(P>0.05)。结论:补肾活血化痰方治疗阿尔茨海默病有效,作用机制与其减少Aβ沉积,促进神经存活的信号传导途径有关。Objective: To observe the effects of Bushen Huoxue Huatan Fang( BSHXHTF) on protein expression of hippocampus extracellular regulated Cdk1 and Cdk2 in rats models with Alzheimer's disease. Methods: The rats models with Alzheimer's disease were established. After 3 days of the model establishment,BSHXHTF were made in to solution with the concentration of 1.5g / ml and then intragastrically administered with the dose of 12 mg / kg. d. The donepezil hydrochloride was intragastrically administered with the dose of 0. 52 mg / kg. d. For the model group and sham operation group were given intragastric administration of normal saline of equal volume,twice per day at 8: 00 and 16: 00 respectively for 4 weeks. The protein expression of extracellular regulated protein kinase in AD rats' hippocampal neurons were observed by immunohistochemical method. Results: The average optical density( AOP) of the positive cells of extracellular regulated Cdk1 and Cdk2 in rats' hippocampal CA1 region was obviously less than the AOP of the sham operation group,and the Cytoplasm staining of most cells are relatively light. The difference was significant( P〈0. 01); while the AOP of the donepezil hydrochloride group and BSHXHTF group increased greatly when compared with that of model group,and some positive cells with much cytoplasm and of light-brown yellowish granule-shape appeared; the difference was significant( P〈0. 01). When the donepezil hydrochloride group and BSHXHTF group were compared with the sham operation group,the color of the former groups were lighter and the AOP of the former group declined significantly;the difference was also significant( P〈0. 01). But the difference was not significant when comparison was made between the donepezil hydrochloride group and BSHXHTF group( P〉0. 05). Conclusion: BSHXHTF has effect on Alzheimer's disease. The action mechanism is related with the decreased Aβdeposit and the promoted signal transduction pathway of nerve survival.
