润肤止痒汤对小鼠慢性湿疹角质形成细胞凋亡及组织病理的影响

摘　　要：目的:探讨"润肤止痒汤"对小鼠慢性湿疹模型角质形成细胞凋亡及组织病理的影响。方法:将80只实验小鼠随机分为4组,每组20只,设为不造模组:正常对照组;造模组:空白对照组、中药治疗组、西药治疗组。造模组60只小鼠耳部应用2,4-二硝基氯苯丙酮溶液小剂量反复刺激制备接触性湿疹模型,中药治疗组:润肤止痒汤汤剂浓煎液,每次灌胃剂量为每只0.75 mg·d-1,日2次,疗程5 d。西药治疗组:盐酸西替利嗪片剂配成水溶液,灌胃剂量为每只0.025 mg·d-1,日1次,疗程5 d。空白对照组给予同等剂量生理盐水灌胃。用免疫组化方法观察各组Bcl-2及Bax蛋白表达情况。结果:小鼠耳部经2,4-二硝基氯苯丙酮溶液最后一次激发后24 h HE病理可见:表皮角化过度,颗粒层细胞轻度增生,棘层细胞增生,上皮角下延,基底层散在液化变性,真皮浅层轻度水肿,间质及血管周围见炎细胞浸润提示造模成功。Bcl-2及Bax蛋白空白对照组表达最明显,中药组与西药组表达低于空白对照组(P<0.05),正常对照组表达低(P<0.05)。结论:在慢性湿疹模型部位促凋亡蛋白大量表达,经中药灌胃治疗后,可降低凋亡通路中促凋亡蛋白表达,调节局部皮损免疫平衡,使局部皮损改善。Objective： To explore the influence of Runfu Zhiyang Decoction on cutin cell apoptosis and histopathology of chronic eczema of mice. Methods： 80 healthy mice were chosen. Among of them,60 mice＇s ears were given repeated simulation with small dose of dinitrochlorobenzene acetone solution to prepare contact eczema model,and the other 20 belonged to the normal control group. Then each group＇s Bc1-2 and Bax protein expression were observed with immunohistochemical method. Results：Bc1-2 and Bax expression of the blank control group were the most obvious; the expressions of the Chinese medicine expression and Western medicine expression were lower than those of the blank control group（ P〈0. 05）; The expressions of the normal control group were low（ P〈0. 05）. Conclusion： After the promotion apoptosis protein expression in location of chronic eczema model was treated with administration by gavage with Chinese medicine,it decreased in apoptotic pathway,which can improve local lesions by adjusting the immune balance of local lesions.
