补肾益气活血方对人骨关节炎软骨细胞增殖及相关细胞因子的干预作用

摘　　要：目的:探讨补肾益气活血方对人骨关节炎软骨细胞增殖及相关细胞因子干预作用,为骨关节炎的治疗提供依据。方法:选取本院90例人关节炎患者进行研究,行软骨细胞分离培养传代,设立空白对照组,IL-1组,补肾益气活血方组各30例,空白对照组细胞不添加任何药物,IL-1组添加含IL-1血清,益气活血方组添加IL-1血清和补肾益气活血方血清,观察各组软骨细胞增殖情况,并且运用逆转录聚合酶链技术检测软骨细胞基质蛋白酶13(MMP-13)和II型胶原的表达情况。结果:补肾益气活血组软骨细胞增殖情况与正常对照组及IL-1组相比,差异有统计学意义(P<0.05);补肾益气活血组MMP-13及Ⅱ型胶原表达情况与其他两组比较,差异有统计学意义(P<0.05)。结论:体外条件下,补肾益气活血药方能够抑制炎性因子IL-1诱导的MMP-13表达,且能对抗MMP-13抑制软骨细胞II型胶原合成,促进软骨细胞的增殖,具有保护软骨细胞的作用。Objective： To explore the intervention role of method of reinforcing kidney qi and promoting blood circulation in regulating human osteoarthritis chondral cell multiplication and correlative cell factors,and provide basis for treating patients with osteoarthritis. Methods： 45 patients with osteoarthritis treated in our hospital were studied. Through dissociated cell subculture of chondral cell,the blank blood serum control group. IL-1 group,and group of IL-1 plus serum containing drug for reinforcing kidney qi and promoting blood circulation were established. Each group＇s chondral cell multiplication was observed; And reverse transcription-polymerase chain technology was used to detect its influence on cellular matrix protease 13 and type II collagen. Results：The group of IL-1 plus serum containing drug for reinforcing kidney qi and promoting blood circulation significally promoted osteoarthritis chondral cell multiplication,stimulated cell regeneration,inhibited too much chondral cell death induced by IL-1. Compared with the normal serum control group and IL-1 group,the differences between them had statistical meaning（ P〈0. 05）.The prescription for reinforcing kidney qi and promoting blood circulation greatly inhibited MMP-13 expression function. The expression of type II collagen was clearly higher than that of the other two groups; The differences between them had statistical meaning（ P〈0. 05）. Conclusion： Under the extrinsic condition,the group of IL-1 plus serum containing drug for reinforcing kidney qi and promoting blood circulation can inhibit MMP-13 expression induced by inflammatory factor IL-1,and inhibit MMP-13 against type II collagen synthesis in chondral cells and promote multiplication of chondral cells and protect chondral cells,which is worth studying in clinical practice.
