摘  要:目的研究薏苡仁酯对胶原诱导性关节炎(CIA)小鼠体内Th17细胞及其细胞因子白细胞介素-17(IL-17A)的作用。方法健康SPF级DBA/1 7~8周雄性小鼠20只,测定造模对照组(经生理盐水治疗后CIA小鼠)、薏苡仁酯组(经薏苡仁酯注射液治疗后CIA小鼠)小鼠脾脏Th17细胞及血清中IL-17变化。应用流式细胞术(FCM)分析小鼠脾脏中Th17细胞的比例,采用酶联免疫吸附试验(ELISA)检测小鼠血清中IL-17A含量。结果经薏苡仁酯注射液治疗后,薏苡仁酯组CIA小鼠脾脏中Th17细胞比例较造模对照组显著降低(1.82%比4.99%,P〈0.05),小鼠血清中IL-17A的含量也明显低于造模对照组[(3.54±0.61)pg/m L比(8.31±0.25)pg/m L,P〈0.05]。结论薏苡仁酯对CIA小鼠Th17细胞具有分化作用。Objective To explore the effect of coixenolide on T helper cell 17(Th17) and its cytokine interleukin-17(IL-17A) in mice with collagen-induced arthritis(CIA). Methods Twenty DBA/1 mice at age of 7-8weeks were used to build CIA model, then they were randomly divided into model group and coixenolide-treated group, with 10 mice in each group. Mice in coixenolide-treated group was injected with 25 ml/kg coixenolide and mice in model group were given injection of equivalent normal saline. The proportion of Th17 cells in the spleen of mice and serum IL-17 A were determined by flow cytometry and enzyme-linked immunosorbent assay(ELISA),respectively. Results The proportion of Th17 in the spleen of mice in coixenolide-treated group was significantly lower than that in model group(1.82% vs 4.99%, P 〈0.05). The serum IL-17 A of mice in coixenolide-treated group was significantly lower than that in control group [(3.54±0.61)pg/m L vs(8.31±0.25)pg/m L,P〈0.05]. Conclusion Coixenolide has inhibitory effect on differentiation of Thl7 cells in CIA mice.
