摘  要:目的研究脑健胶囊对大鼠脑组织Fas凋亡途径的影响,探讨脑健胶囊抗脑缺血后神经保护的作用机制。方法将SD大鼠随机分为补阳还五汤组、脑健胶囊组、模型组和假手术组共4组,每组25只。除假手术组和模型组给予等体积无菌蒸馏水以外,其余两组在大脑中动脉线检法复制局灶性脑缺血大鼠模型(MCAO)造模后给药,将各组大鼠分别存活1、3、7、14、28 d时取脑组织及腹主动脉采血。运用免疫组化法和Elisa法检测脑组织及血清中Fas相关死亡域蛋白(FADD)、Caspase-8、Caspase-3因子表达。结果脑缺血后大鼠均有神经功能障碍,经过治疗后,脑健胶囊能显著改善其神经功能缺失症状,与模型组比较,差异具有统计学意义(P<0.05)。结论脑健胶囊通过抑制缺血后脑组织FADD、Caspase-3、Caspase-8的表达,从而起到保护缺血后脑组织作用,减轻脑组织损伤。Objective To study the effect of Naojian capsule on Fas apoptotic pathway in brain tissues of rats and discuss its neuroprotective effect on anti-cerebral ischemia. Methods SD rats were randomly divided into 4 groups, including Buyang Huanwu Decoction group, Naojian Capsule group, model group and sham-operation group, 25 rats in each group. Isopyknic sterile distilled water was garaged in sham-operation group and normal group; Other groups were given medicine after molding the focal MCAO rats by suture method. Blood sampling in aorta abdominalis and brain tissue were collected at 1, 3, 7, 14, 28 d of each group. The expressio ns of FADD, Caspase-8 and Caspase-3 in cerebral tissue and blood serum were detected by immunohistochemical and Elisa methods. Results All of the rats showed obvious neurological function damage after cerebral ischemia, and neurological deficit were improved obviously by Naojian capsule. Compare with the model group, there is statistical significance (P〈0.05). Conclusion Naojian Capsule exhibited the protective cerebral tissues effect in rats with cerebral ischemia by inhibiting expressions of FADD, caspase-3 and caspase-8, and relieved the damage.
