“补肾活血”针法对快速老化模型小鼠SAMP8学习记忆能力及海马CA1区NEP表达的影响

摘　　要：目的研究"补肾活血"针法对快速老化模型小鼠SAMP8(Senescence Accelerated Mouse Prone 8,SAMP8)学习记忆能力及海马CA1区中性内肽酶(neprilysin,NEP)表达的影响。方法将6月龄雄性快速老化模型小鼠SAMP8 20只随机分为针刺组和SAMP8对照组,各组10只;另选取10只同龄雄性正常老化小鼠SAMR1(Senescence Accelerated Mouse/resistant 1)为正常对照组。针刺组针刺"百会"、双侧"血海"、"肾俞"以及"膈腧"4周。治疗结束后,采用Morris水迷宫测定"补肾活血"针法对小鼠学习记忆能力的影响,免疫组化方法检测小鼠海马CA1组织中NEP的表达情况。结果针刺组学习记忆能力明显较SAMP8对照组增强(潜伏期缩短、跨平台象限次数增加)(P<0.05);针刺组NEP相对表达量明显较SAMP8对照组增高(P<0.05)。结论"补肾活血"针法可能通过上调NEP表达,促进Aβ降解,改善Aβ相关的认知障碍来防治阿尔茨海默病,这可能是"补肾活血"针法防治阿尔茨海默病作用机制的一部分。Objective To study the effect of ＂Bushen Huoxue＂ acupuncture on the ability of learning and memory and the expression of NEP in the hippocampal CA1 region of the rapid aging SAMP8 mouse models. Methods Six-month aging SAMP8 male mice were divided into the acupuncture group and control group, 10 in each group, and 10 normal aging SAMR1 male mice with the same age were selected as normal control group. The acupuncture group was treated by acupuncture at Baibui acupoint, bilateral acupoint of Shenshu and Xuehai, and Geshu acupoint for 4 weeks. At the end of the course of treatment, the abilities of learning and memory were tested by Morris water maze and the expression of NEP in hippocampal CA1 region were detected by Immunohistochemical method. Results The abilities of learning and memory in the acupuncture group （the shorter escape latency and more times in platform quadrant） were significantly increased than that in SAMP8 control group （P〈0.05）. The NEP expression levels in the acupuncture group were higher than that in SAMP8 control group （P〈0.05）. Conclusion ＂Bushen Huoxue＂ acupuncture may prevent Alzheimer＇s disease by increasing the NEP level, accelerating AI3 degradation, then improving A[3 related cognitive disorder. That mechanism may be a portion of ＂Bushen Huoxue＂ acupuncture on treating Alzheimer＇s disease.
