摘 要:本试验基于酶标仪法建立了用于快速、准确检测牛奶中硫氰酸根浓度值的方法,该方法用三氯乙酸除去牛奶中的蛋白和脂肪,过滤后取上清液,硫氰酸根与硝酸铁溶液显色后,用酶标仪在450 nm波长处测吸光度值,用模拟奶做标准曲线,通过科学的数据处理,定量测定牛奶中硫氰酸盐的含量。试验检测体系适用对象为生鲜乳及复原乳,试验结果表明:当硫氰酸根的浓度在0.5~16 mg/kg范围内时,硫氰酸根的质量浓度与溶液的吸光度值呈现良好的线性关系,相关系数为0.999 7,回收率为98.6%~105%,仪器检测下限为0.3 mg/kg。本试验所建立的方法数据可信度高,检测灵敏度高,成本低廉,操作方便,适合中小单位对牛奶中硫氰酸根做定量分析。In order to determine the concentrantion of SCN-in the milk, we have developed a method with the microplate reader . We cleared the protein and fat in the milk with trichloroacetic acid,based on the reaction between Fe3 + and different concentration of SCN-, different absorbance value would occur. Determining the concentration of SCN- with the standard curve which was made with simulated milk was available. Result :The results showed that the system could be used to determine raw milk and reconstituted milk .In addition ,the result showed that the concentration of SCN-between 0.5 mg/kg and 16 mg/kg formed a good Linearship with absorbance (R^2=0.999 7). The recovery was between 98.6%-105%. Conclusion: The method was accurate,reliable and simple without using expensive devices.
