摘 要:目的研究呼吸氢气对支气管哮喘(简称哮喘)大鼠模型的治疗作用,并初步探讨其可能的作用机制。方法建立大鼠卵蛋白哮喘模型,之后对哮喘模型动物进行氢气吸入治疗。治疗1周后,通过气道阻力测定和肺组织形态学观察评价治疗效果,并进一步通过检测氢气对肺组织内丙二醛(MDA)水平的影响,探讨可能的氢气作用机制。结果哮喘模型组气道阻力明显高于对照组(P〈0.01),氢气吸入治疗可有效抑制卵蛋白激发引起的气道阻力升高(P〈O.05)。卵蛋白诱发明显的肺组织病理学改变,表现为细、小支气管的管壁和伴行动脉周围有较多嗜酸细胞、单核细胞、浆细胞和淋巴细胞浸润,气道、血管平滑肌及肺泡壁增厚。而氢气治疗可有效缓解肺组织病理学改变。哮喘模型组动物肺组织内MDA水平显著升高(P〈O.01),氢气吸入治疗可有效阻断卵蛋白引起的肺内MDA升高(P〈O.01)。结论吸入氢气可有效治疗卵蛋白引起的大鼠哮喘,机制可能是通过氢气的抗氧化作用。Objective To study the therapeutic effects of hydrogen inhalation on asthmatic rat model,and the possible underlying mechanisms. Methods The asthmatic model was established by ovalbumin injection and inhalation. Then the asthmatic rats were treated with hydrogen. One week later, airway resistance determination and histomorphological experiments were performed to evaluate the protective effects of hydrogen on bronchial asthma (asthma). Further study the underlying mechanisms by detecting the levels of MDA in the lung tissues. Results The airway resistance of asthmatic group significantly increased compared with those of control group (P 〈0.01). Moreover, treatment with hydrogen for 1 week resulted in blocking the increase of airway resistance ( P 〈0.05). Lung damage was assessed based on HE staining. OVA-induced rats showed extensive infiltration by inflammatory cells around airway and accompanying artery, including acidophilicgranulocytes, monocytes, phlogocytes and leukomonocytes. The airway, vascular smooth muscle and alveolar walls were thick induced by OVA. While these effects were not observed in control animals. After hydrogen treatment, histological symptoms of asthma were much improved. OVA induced a significant increase of MDA in the lung tissue ( P 〈 0.01). Hydrogen treatment blocked the increase of MDA induced by OVA ( P 〈0.05). Conclusions Inhalation hydrogen has protective effects on asthma induced by OVA, and the possible mechanism is antioxidant property of hydrogen.
