乌梅丸对溃疡性结肠炎实验大鼠血清IL-6和IL-10水平的影响

摘　　要：目的:观察乌梅丸对溃疡性结肠炎(ulcerative colitis,UC)实验大鼠血清白细胞介素-6(interleukin-6,IL-6)和白细胞介素-10(interleukin-10,IL-10)水平的影响,探讨其治疗UC的作用机制。方法:雄性SD大鼠采用2,4,6-三硝基苯磺酸乙醇法复制UC模型。造模成功后,随机均分为正常组、模型组、柳氮磺胺吡啶组和乌梅丸组,各组12只。造模后第3天开始,实验动物灌胃给药,连续给药10 d,第14天腹主动脉采血取血清,用ELISA法测定血清IL-6、IL-10水平;解剖观察大鼠结肠黏膜损伤指数。结果:乌梅丸明显改善结肠组织的病理损伤,乌梅丸组与模型组比较,血清IL-6水平显著减低,IL-10水平升高(均为P<0.01)。乌梅丸组与柳氮磺胺吡啶组比较,血清IL-6,IL-10水平变化差异无统计学意义(均为P>0.05)。结论:乌梅丸可能增加抑制促炎细胞因子,减少抗炎细胞因子生成,从而发挥对UC的治疗作用。Objective： To observe the effect of Wumei Pill on interleukin-6（ IL-6） and interleukin^-10（ IL-10） levels of serum of ulcerative colitis in rats,to explore the therapeutic mechanism of ulcerative colitis（ UC）. Methods： Male SD rats,using 2,4,6-trinitrobenzene sulfonic acid alcohol method replicates UC ethanol method. After the success of modeling,they were randomly divided into normal group,model group,sulfasalazine（ SASP） group and Wumei Pill group,with 12 mice in each group. Third days after modeling experimental animal began intragastric administration,continuous administration of 10 d for fourteenth days,abdominal aortic blood serum,serum IL-6,IL-10 levels were determined by ELISA method. Observed the colon mucosal injury. Results： Wumei Pill significantly improves the pathological injury of colon tissue. Compared with model group,the serum IL-6 level of Wumei Pill group was significantly decreased,IL-10 levels increased（ P〈0. 01）. Comparison of Wumei Pill group and SASP group,the serum levels of IL-6 and IL-10,there was no significant difference（ P〉0. 05）. Conclusion： Wumei Pill can inhibit proinflammatory cytokines,promote the formation of antiinflammatory cytokines so as to play a therapeutic effect on UC.
