摘  要:目的研究低剂量长期天然放射性高本底辐射地区(HBRA)人群的周围血线粒体细胞色素C氧化酶亚基Ⅱ(COXⅡ)基因表达水平。方法随机选择HBRA和对照地区(CA)各50名年龄为50.0—59.0岁男性居民作为HBRA组和CA组。采集2组人群肘静脉血分离白细胞和血浆,实时荧光定量聚合酶链式反应测定白细胞中COX1ImRNA相对表达水平,酶联免疫吸附实验测定血浆中COXⅡ蛋白水平。结果HBRA组人群白细胞COXⅡmRNA相对表达水平高于CA组[(13.01±1.76)vs(1.00±0.23),P〈0.01],HBRA组人群血浆中coxI蛋白水平与CA组比较,差异无统计学意义[1.10(0.58,1.56)vs0.88(0.44,1.57)mg/L,P〉0.05]。结论低剂量长期天然放射性高本底辐射能导致线粒体COXⅡ基因表达水平上升,可能与适应性反应机制相关。Objective To examine the expression level of mitochondria eytochrome C oxidase Ⅱ ( COX Ⅱ) among the peripheral blood leukocytes of inhabi-tants in high background radiation area (HBRA) in Guangdong, China. Methods Fifty male inhabitants aged from 50.0 to 59.0 in HBRA and the other 50 inhabitants in control area (CA) were randomly selected as HBRA group and CA group respectively. The fasting blood was taken from cubital vein of each subject, and the white blood cells and plasma serum was segregated for detection. Real-time fluorescence quantitative PCR was applied to determined the mRNA relative expression level of COX U in the white blood ceils. Concentration of COX Ⅱ in plasma was also determined with the method of ELISA. Results The mRNA relative expression level of COX Ⅱ in HBRA group was higher than that of the CA group [ ( 13.01 ± 1.76) vs ( 1.00 ±0.23), P 〈 0.01]. While the concentration of COX Ⅱ in plasma in HBRA was lower than that of CA group [1. 10(0.58, 1.56) vs 0.88(0.44, 1.57) mg/L, P〉0.05]. Conclusion Expression level of COX Ⅱ of mitochondria may be enhanced under the long-term natural high background ionizing radiation, which may be related to the mechanism of adaptive response.
