摘  要:目的:观察辛热药附子对薄荷油诱导大鼠虚寒状态的影响,以期为辛热药药性研究提供实验依据。方法:将大鼠随机分为正常组,虚寒组,附子大、中、小剂量组,持续造模21d,于造模第15天开始灌服各组药物,连续7d,在第22天,取血,检测内分泌系统、免疫系统、物质代谢、一氧化氮(NO)/环磷酸鸟苷(c GMP)等相关指标。结果:附子各剂量组均不同程度地表现出一定的治疗调节作用,尤以大剂量效果最好,主要体现在对动物生长情况(体质量、外观等)、内分泌系统功能(尤其甲状腺功能T3、T4、TSH)、免疫系统功能及部分物质代谢水平方面的影响,附子大剂量对NO/c GMP也均有调节作用。结论:附子可能通过NO/c GMP这一生物机制调节下丘脑的神经内分泌功能,从而纠正虚寒状态下的甲状腺功能、性腺功能、免疫功能、物质代谢水平的异常,体现其辛热药性。Objective: To provide an experimental evidence for the pungent-hot herb property research and to exemplify the application of this model in drug research, based on the initially successful modeling of rat's deficiency-cold situation induced by spearmint oil, Radix Aconiti Lateralis Preparata, as a representative pungent-hot herb, to interfere and observe index's changes of deficiency-cold. Methods: The rats were randomly divided into normal group, deficiency-cold group, large, medium and small doses groups. The modeling lasted for 21 days. From the 15 th day, each group was gavaged with drug for 7 days, and the blood was drawn at the 22 nd day. The endocrine system, immune system, metabolism, nitric oxide(NO)/cyclic guanosine monophosphate(c GMP) and other relevant indicators were detected. Results: Radix Aconiti Lateralis Preparata in each group showed regulation function to different degrees, especially the large dose group. The results were mainly reflected in the growth of animals(weight, appearance, etc), endocrine system function(especially thyroid function indexes: T3, T4, TSH), immune system function and substance metabolism effects. The large dose group also had regulation function in NO/c GMP. Conclusion: Radix Aconiti Lateralis Preparata would regulate the function of hypothalamic neuroendocrine through a possible biological mechanism of NO/c GMP to adjust the thyroid function, gonad function, immune function and metabolism abnormalities of deficiency-cold which reflected its pungent-hot herb property.
