舒筋活血止痛水外擦拍打对兔膝骨性关节炎软骨组织MMP-1表达的影响

摘　　要：目的:观察舒筋活血止痛水外擦拍打治疗对兔膝骨性关节炎(KOA)软骨组织MMP-1表达的影响。方法:随机将15只新西兰大白兔分为正常组、模型组、拍药组,采用前交叉韧带切断术(ACLT),制作KOA模型。术后6周拍药组进行舒筋活血止痛水外擦拍打干预。干预2周后处死动物,采用HE染色观察软骨形态学改变,并进行改良Mankin评分,免疫组化检测MMP-1的表达情况。结果:模型组改良Mankin评分明显增高,拍药组改良Mankin评分低于模型组,差异有统计学意义(P<0.01);模型组MMP-1阳性表达显著增强,拍药组MMP-1阳性表达较模型组降低,差异有统计学意义(P<0.01)。结论:舒筋活血止痛水外擦拍打治疗可明显降低兔KOA软骨组织MMP-1的表达,延缓兔KOA的进展。Objective： To explore the effect of shujin huoxue zhitong shui on rabbits knee osteoarthritis about the chondrocytes MMP- 1 expression. Methods： Fifteen rabbits were randomly divided into three groups： normal group, model group and drug group. In addition to the normal group, the other two group rabbits received anterior cruciate ligament transaction, and the interven- tion carried out after six weeks. Remove the femoral condyle cartilage after two weeks intervention. Hematoxylin-eosin staining was conducted for histological observation of rabbit articular cartilage, and the cartilage structure were assessed and quantified with the modified Mankin rating criteria; immunohistochemistry were used to assess the expressions of MMP-1 in rabbit chondrocytes. Re- sults：The modified Mankin score is significantly higher of model group, the score of drug group is lower than model group, with a significant difference （P〈0.01）. There is a strong expression level of MMP-1 in the model group rabbit chondrocytes. The MMP- 1 expression level is lower in drug group, with a significant difference （P〈0. 01）. Conclusion： Shujin huoxue zhitong shui can de- lay the development of osteoarthritis by inhibiting the MMP-1 expressing of rabbit ehondrocytes.
