摘  要:以药用真菌红芝(Ganoderma lucidum)为出发菌,通过紫外线(Ultraviolet ray,UV)、亚硝基胍(Nitrosoguanidine,NTG)及紫外线(UV)-亚硝基胍(NTG)逐级诱变处理其菌丝体片段,采用平板变色法初筛、愈创木酚法测定发酵液漆酶酶活力复筛,获得1株漆酶高产诱变菌株LYL263。用复筛液体培养基震荡培养时,其平均酶活力比出发菌株提高187%,达到145 500 U/L,且产酶稳定。The mycelium fragments of Ganoderma lucidum were exposed successively under ultraviolet ray, nitrosoguanidine, ultraviolet ray-nitrosoguanidine and the mutants were screened in guaiacol-containing SPDA plates, and then the laccase activities of the mutated strains were determined by guaiacol. A mutant LYL263 with high yield of laccase was obtained. The average activity of laccase from LYL263 had increased up to 145 500 U/L, and its ability of producing laccase was stable on culture condition of liquid medium with glucose and yeast extract.
