摘 要:目的研究新生期大鼠间断、重复吸人七氟烷,对其成年期行为学及β淀粉样蛋白(Aβ)、异常磷酸化Tau蛋白(PHF—Tau)在海马CA1区的表达。方法7日龄SD大鼠24只,随机分为3组(n=8),1.5%七氟烷组(S1组),2.6%七氟烷组(S2组),对照组(C组)。分别于出生7、14、21d(P7、P14、P21)吸入1.5%、2.6%七氟烷及运载气体1h,于P91~P95行定位航行和空间探索实验。水迷宫实验结束后,取海马组织,用免疫组织化学的方法测定CA1区Aβ、PHF—Tau蛋白的表达。结果①与C组相比,P94,S1组、S2组逃避潜伏期延长(P〈0.01或P〈0.05),P95,S1组潜伏期延长(P〈0.05);②与P91相比,C组、S2组P94和P95逃避潜伏期缩短(P〈0.05);S1组,P93逃避潜伏期缩短(P〈0.05);③与C组比较,s1组原平台象限滞留时间百分比减少(P〈0.01);④与C组比较,S1组PHF—Tau蛋白MOD增加(P〈0.05),S1组、S2组Ap蛋白MOD增加(P〈0.05或P〈0.01)。结论新生期大鼠间断、重复吸入1.5%、2.6%的七氟烷可使其成年期学习记忆能力下降,其中吸入1.5%七氟烷较2.6%七氟烷影响更持久。其机制可能与成年期海马CA1区AB、PHF-Tau蛋白水平的改变有关。Objective To study the effects of interrupted and repeated inhalation of sevoflurane in neonatal rats on praxiology of adulthood and the expressions of amyloid beta - protein (Aβ) and abnormal phosphorylated Tau ( PHF - Tau) protein in the CA1 area of hippocampus. Methods Twenty - four Seven - day old neonatal SD rats were divided into 3 groups randomly (n = 8 in each group), 1.5% sevoflurane (group S1 ), 2.6% sevoflurane (group S2 ) and the control group ( group C). Rats in each group were inhalated 1.5%, 2.6% sevoflurane and carrier gas for 1 h in the 7th( P7 ) , 14th(P14), 21th day (P21), respectively. Rats were tested via a water maze experiment in P91 -P95 and the escape latency and retention time in the original platform quadrant and the per- centage of entering times in spatial probe test were recorded. The hippocampus afer morris water maze test was collected to determine the expressions of Aβ and PHF - Tau protein with immunohistochemical method. Results 1 )Compared with group C, escape latencies in groupS1 and group S2 were prlonged (P 〈 0.01 or P 〈 0.05 ) in the 94th day and in group S1 in the 95th day (P 〈0.05) ;2)The escape latency was decreased in P94 and P95 of group C and group S2 and P93 of group S1 compared with that in P91 (P 〈 0.05 ) ;3 )Compared with group C, the percentage of retention time in the original platform quadrant was significantly reduced in group S1 (P 〈 0.01 ) ;4) Compared with group C, the mean opticaldensity (MOD) of PHF - Tau protein in group S1 (P 〈0.05 ) and Aβ in group S1 and S2 (P 〈 0. 01 or P 〈 0.05 ) was increased. Conclusion The interrupted and repeated inhalation of 1.5% and 2.6% sevoflurane on neonatal rats could impair learning and memory in adulthood, in which the effects of inhalation of 1.5% sevoflurane lasted longer than those of 2.6% sevoflurane. The mecha- nisms might be related to the deposition of Aβ protein and changes of the PHF- Tau protein expression.
